半夏泻心汤对幽门螺杆菌致人胃黏膜上皮细胞GES-1凋亡的影响

摘　　要：[目的]观察半夏泻心汤对幽门螺杆菌致人胃上皮细胞GES-1调亡的干预作用。[方法]实验分正常组、感染组和半夏泻心汤组,感染组和半夏泻心汤组按50∶1的细菌∶细胞比例感染GES-1细胞,正常组和感染组均以正常鼠血清进行培养,半夏泻心汤组以含药鼠血清进行培养,采用MTT法检测细胞的增殖情况,DAPI染色分析细胞凋亡情况。[结果]与正常组比较,感染组的细胞增殖显著下降(P<0.05),细胞凋亡明显提高(P<0.05)。与感染组相比,10%、15%的含药血清能促进细胞增殖(P<0.05),半夏泻心汤组能降低细胞凋亡(P<0.05)。[结论]半夏泻心汤具有改变幽门螺杆菌对胃黏膜上皮细胞凋亡的作用。[Objective]To observe the effect of banxia xiexin decoction on apoptosis of human gastric epithelial cell GES-1 infected by Hp[.Methods]In the study,3 groups were applied,including control group,infected group and banxia xiexin decoction group.GES-1 cells were infected at bacteria/cells ratio of 50∶1 in the last two groups.Cells were cultured in normal rats’serum in the first two groups,and those were cultured in medicated rats’serum in banxia xiexin decoction group.Cell proliferation was determined by MTT assay,and apoptosis was observed through DAPI staining analysis.[Results]Compared with the control group,cell proliferation greatly increased（P 0.05）,while apoptosis significantly decreased in infected group（P 0.05）.Compared with infected group,apoptosis decreased in banxia xiexin decoction group（P 0.05）,and medicated serum at concentration of 10% and 15% promoted proliferation（P 0.05）.[Conclusions]Banxia xiexin decoction can change the effect of Hp on apoptosis of gastric epithelial cells.
