摘 要:目的:研究三七和栀子有效成分对APP转基因小鼠脑内淀粉样蛋白的影响。方法:转基因小鼠雌雄各半,分别随机分为三七和栀子有效成分高剂量组、中剂量组、低剂量组,阳性药安里申组,模型组。同窝出生的野生型小鼠作为阴性对照组。小鼠自6月龄开始自主进食给药4个月,然后进行免疫组织化学、Western Blot、ELISA实验,检测海马和皮层区淀粉样蛋白Aβ1-40、Aβ1-42及APP的表达水平和脑内的淀粉样斑块。结果:在雌性小鼠各组中,三七和栀子有效成分低剂量组能够显著性降低海马区Aβ1-40和Aβ1-42的水平,且淀粉样蛋白前体蛋白APP水平显著性降低。结论:三七和栀子有效成分可能作用于APP生成阶段,通过下调APP的产生,从上游降低了Aβ产生的风险。Objective: To investigate the effect of the active ingredients from Sanchi and Gardenia on β-amyloid protein of the APP transgenic mouse brain. Methods: Transgenic mice were divided into halves: male and female. And each group was randomly divided into high dose, middle dose, low dose of herb treatment groups, positive control and model groups. Negative control group contain wild-type mice which are littermates of transgenic ones. The expression levels of Aβ1-40, Aβ1-42, APP in hippocampus and cortex were detected by Western Blot and ELISA tests, while amyloid plaques in brain were examined using immunohistochemistry methods. Results: In female group, Sanchi and Gardenia low dose group shows significantly reduced levels of Aβ1-40, Aβ1-42 in hippocampus as well as APP protein. Conclusion: The active ingredients of Sanchi and Gardenia may have the effect on the expression level of APP, reducing the risk of Aβ production from the upstream by down-regulating the expression level of APP.
