摘 要:放线菌GR54对油茶叶枯病具有良好的拮抗效果。为对其进行开发利用,对其发酵条件的优化进行研究,以达到对油茶主要病害的防治作用。研究不同发酵培养液、碳氮源等营养因子,以及培养时间、接种量、装液量、初始pH值等非营养因子对放线菌发酵液抑菌活性的影响。综合考虑菌体干重和对油茶叶枯病菌的抑菌率2个指标,生防放线菌的优化发酵培养液为葡萄糖15 g、大豆粉10 g、NaCl 5 g、酵母膏1 g、CaCO31 g、蒸馏水1 000 mL,pH值7.0,接种量10%,装液量100 mL,28℃、160 r/min振荡培养7 d。生防放线菌GR54的发酵培养液可有效抑制油茶叶枯病病原菌,抑制率可达93.05%。Actinomycete GR54 has a good ability against Camellia oleifera leaf blight.A lots of researches on the optimization of culture fermentation conditions of actinomycete GR54 against Camellia oleifera leaf blight were conducted in order to control the disease.The effects of trophic factors(different fermentation culture solution,carbon and nitrogen sources) and non-trophic factors(incubation time,inoculum size,loaded liquid and initial pH value) on the antibacterial activity of the fermentation broth were investigated.By take two factors(mycelium dry weight and rate of inhibiting bacteria) into consideration,the best cultural solution formula consisted of glucose 15 g,soybean meal 10 g,NaCl 5 g,yeast extract 1g,CaCO3 1g,distilled water 1 000 mL,pH7.0,inoculum size 10%,loaded liquid 100 mL,28℃,160 r/min shaking cultivation for 7 days.The fermentation broth of the GR54 can effectively inhibit the pathogenic bacteria of Camilla oleifera leaf blight with an inhibitory rate of 93.05%.
