摘  要:目的:研究穴位电针对大鼠下颌前伸后咀嚼肌中基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)表达的影响。方法:60只雄性SD大鼠随机分为12组(n=5):空白对照3、7、14、21 d组,条件对照3、7、14、21 d组和实验3、7、14、21 d组。采用免疫组化染色法和荧光定量PCR法检测大鼠二腹肌前腹中MMP-9表达的差异。结果:与空白对照组比较,条件对照组在7、14 d时MMP-9表达明显上升(P<0.05),实验组在3、7、14 d时MMP-9的表达明显上升(P<0.05);与条件对照组比较,实验组在3、7 d时MMP-9的表达明显上升(P<0.05),14 d时两组无明显差异(P>0.05),实验组21 d MMP-9表达量降低(P<0.05)。结论:穴位电针可早期上调大鼠下颌前伸咀嚼肌中MMP-9的表达,促进大鼠咀嚼肌的适应性改建。Objective:To investgate effects of acupoints electroacupuncture on the expression of matrix metalloproteinase-9(MMP-9) in the mandibular protrusion of rats.Methods:Totally 60 male SD rats were randomly divided into 4 blank control groups,4 conditional control groups and 4 experimental groups(n=5,respectively).Rats were sacrificed on 3,7,14,28 d after experimental mandibular protrusion.Immunohistochemistry and real-time PCR methods were used to detect expressions of MMP-9 in growing rats'masticatory muscle.Results:Compared with those in blank control groups,MMP-9 expressions were significantly increased in conditional control group on 7,14 d,with statistical differences(P〈0.05);MMP-9 expressions were significantly increased in experimental group on 3,7,14 d,with statistical differences(P〈0.05).Compared with those in conditional control group,MMP-9 expressions were significantly increased in experimental group on 3,7 d,with statistical differences(P〈0.05);there was no significant difference between the two groups on 14 d(P〈0.05);MMP-9 expressions were significantly decreased in experimental group on 21 d,with statistical differences(P〈0.05).Conclusions:Electric acupuncture can up regulate the expression of MMP-9 in rats masticatory muscle after functional mandibular protrusion,which may improve the treatment effect of functional appliance.
