摘 要:采用暴露实验方法,研究了不同浓度HCB(分别为0.0、2.0、4.0、8.0、16.0 mg/L)在不同暴露时间(0-14d)下对梨形环棱螺肝脏和鳃中MDA、ROS含量以及DNA单链断裂程度的影响。结果表明:HCB对梨形环棱螺肝脏和鳃中MDA、ROS含量和DNA断裂均有明显影响。肝脏和鳃的MDA含量在HCB暴露过程中波动升高,直到在第4天或第7天达到峰值,然后MDA含量持续下降,到第14天时,除了16.0mg/L剂量组与对照组无显著差异外,其他剂量组MDA仍处于显著被诱导状态。ROS水平随暴露时间和剂量增加而表现出升高的趋势,并且高剂量组较中低剂量组更早达到峰值,随着暴露时间的进一步延长,各剂量组ROS含量均有一定程度的下降,但在实验结束时仍然显著高于对照组。各剂量组DNA在HCB暴露起始时,出现一定程度的单链断裂现象,即F值迅速降低,随后F值有所上升,在第4天时F值达到最高点,显示DNA有一定程度的修复效应。接着F值呈现出持续下降的趋势,并且高剂量组的DNA单链断裂明显,表现出时间-剂量效应。结果提示,上述生物标志物在六氯苯对梨形环棱螺进行毒性暴露过程中的变化较为敏感,可以作为指示六氯苯对梨形环棱螺毒理效应的生物标志物。An exposure experiment was conducted to study the effects of HCB at different concentrations (0, 2.0, 4.0, 8.0, 16.0 mg / L) on levels of MDA and ROS as well as the extent of DNA single-strand breaks in liver and gills of Bellamya purificata at different exposure time (0-14d) in vivo. The results showed that HCB had significant influence on them. The concentrations of MDA increased fluctuatly during exposure of HCB until they reached peak value on 4th day or 7th day, and then they decreased on 14th day. There was significant difference between HCB exposure and the control group except 16.0 mg/L HCB group. The concentration of ROS tended to be enhanced with the exposure time and dosage increasing, ROS of high dosage group reached its peak earlier than that of low dosage. The content of ROS decreased in a certain extent with the exposure time further increasing, and they were still significantly higher than that of control group on 14th day. DNA single-strand breaking of each HCB group occurred in a way at the beginning of exposure, that was the F value decreased quickly, and then increased slightly till it reached an upper value on 4th day, which indicated that there were some recovering of the breaking DNA. Subsequently, F value of all HCB groups keeped decreasing, and more breaking appeared in high-dose-HCB groups. There was a time-dosage effect between the HCB exposure and DNA single-strand breaking. All these results suggested that biomarkers in this study were sensitive during HCB exposuring, which could be used as biomarkers of evaluating toxicological effects of HCB on B. purificata.
