摘  要:目的:观察急性高眼压后大鼠视网膜葡萄糖转运因子-1(GLUT-1)的表达变化及其对视网膜损伤的影响。方法:利用急性高眼压大鼠模型,通过免疫组织化学和免疫印迹检测急性高眼压后大鼠视网膜GLUT-1蛋白的表达变化;Nissl染色观察视网膜神经细胞层次改变及节细胞数的变化。结果:正常视网膜血管内皮细胞有GLUT-1表达,急性高眼压后3h开始下调,6h达最低值,之后1d、3d、7d逐渐恢复至正常水平。Nissl染色结果显示急性高眼压后3h、6h和12h视网膜节细胞数减少不明显,到1、3d,视网膜层次紊乱、变薄,节细胞数明显减少。结论:急性高眼压后早期大鼠视网膜GLUT-1表达下调,影响了视网膜葡萄糖的转运,这可能是视网膜结构紊乱,节细胞丢失的重要原因。Objective: To observe the changes of glucose transporter-1 (GLUT-l) expression in rat retina and its effect on the retina following the rat acute high intraocular pressure (AHIOP). Methods. The rat model of AHIOP was established, and the changes of GLUT-1 protein was detected by immunohistochemistry and Western blotting. Nissl staining was used to ob- serve the changes of the retina structure and the reduction of retinal ganglion cells. Results: Immunohistochemistry and West- ern blotting showed that GLUT-1 was expressed in the endothelial ceils of the normal retina, began to decrease at 3 h, reached the lowest value at 6 h, and gradually recovered to normal on 1 d, 3 d and 7 d. Nissl staining showed that the layers of the retina was gradually being disorder at 3 h, 6 h and 12 h; while the loss of the retinal ganglion ceils was not obvious. The outer nuclear layer, inner nuclear layer and ganglion cells layer were being further disorder and thinner, and the number of retinal ganglion cells was significantly reduced on 1 d and 3 d. Conclusion: The down-regulation of GLUT-1 at the early stage after AHIOP in rat retina affects the retinal glucose transport, which may be an important factor of the retinal structural disorder and ganglion cell loss.
