大鼠肝纤维化过程中ACE 2、Ang(1-7)的动态变化

摘　　要：目的探讨大鼠肝纤维化形成过程中ACE2、Ang(1-7)的变化情况。方法成年雄性Wistar大鼠36只,随机取6只为正常对照组,余30只制备CCl4诱导大鼠肝纤维化模型,分别在造模第15、30、45、60、75天随机处死6只,对各组大鼠肝组织进行HE及Masson染色观察其病理变化,ELISA检测肝匀浆Ang(1-7)的水平,免疫组织化学方法检测肝组织ACE2蛋白的表达,Real-timePCR检测ACE2及TGF-β1mRNA的表达。结果随着大鼠肝纤维化进展,肝组织中ACE2、Ang(1-7)、TGF-βl表达水平呈上升趋势(P<0.01),但第60天至75天Ang(1-7)增加不明显,ACE2的表达水平下降(P<0.05)。结论 TGF-β与肝纤维化程度正相关,ACE2、Ang(1-7)参与肝纤维化的发生发展。Objective To observe the changs of ACE 2;Ang （1-7） in rat liver fibrosis. Methods From 36 adult male Wistar rats, six were randomLy selected as the normal control group, and the remaining 30 were given subcutaneous injection of 40%chronic carbon tetrachloride （CCl 4） to induce liver fibrosis. Six were killed, respectively, in the modeling days of 15, 30, 45, 60 and 75. Histopathological study of liver tissue was done with hematoxylin-eosin （HE） and rapid Masson staining. Enzyme-linked immunosorbent assay（ELISA） was used to determine the levels of Ang（1-7） of liver homogenates. The expression of ACE 2 protein was evaluated by immunohistochemistry. Real-time PCR was used to detect the expression of ACE 2 mRNA and TGF-β1 mRNA. Results With the rat liver fibrosis progression in liver tissue, the levels of ACE 2, Ang（1-7）, TGF-β1 showed an upward trend in the 60 days prior to the modeling（P〈0.01）. Ang（1-7） did not significantly increase from 60 days to 75 days, while the expression of ACE 2 decreased（P〈0.05）. Conclusion TGF-β1 is positively correlated with liver fibrosis, ACE 2、Ang （1-7） participate in the development of liver fibrosis.
