摘 要:目的观察螺旋藻多糖(PSP)对乙酸烧灼型胃溃疡大鼠血清中活性成分的影响,初步探讨其治疗胃溃疡的可能机制。方法采用乙酸烧灼法制备实验性胃溃疡大鼠模型,将50只Wistar大鼠随机分成正常组、模型组、奥美拉唑组和PSP高、低剂量(400、200 mg/kg)组,每组各10只,连续药物治疗15 d后取血,分别采用硝酸还原酶法、黄嘌呤氧化酶法及硫代巴比妥酸法测定大鼠血清中一氧化氮(NO)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果 PSP高低剂量组与奥美拉唑组均能升高血清NO含量[(26.71±6.47)、(32.60±4.65)μmol/mL],提高血清SOD活力[(292.74±52.44)、(303.57±13.80)U/mL],降低MDA含量[(15.60±2.39)、(15.57±2.19)nmol/mL],与模型组比较差异有统计学意义(P<0.05或P<0.01)。结论 PSP具有一定的抗胃溃疡的作用,其机制可能与提高血清NO含量和SOD活性、降低MDA含量有关。Objective To investigate the effect of PSP on serum active ingredients in rats with experimental gastric ul- cer, and to make a preliminary discussion on the possible mechanism of it in treating gastric ulcer. Methods The model of experimental rats with gastric ulcer was made by acetous cauterization method. 50 Wistar rats were randomly separated into normal group, model group, Omeprazole group, and high and low dose (400, 200 mg/kg) of PSP group with 10 rats in each group. Blood was collected after therapies for 15 days. NO, SOD, MDA levels were measured by nitrate reductase activity, xanthine oxidase and thiobarbituric acid measurement. Results High dose of PSP group and Omeprazole group could increase the content of NO [(26.71±6.47), (32.50±4.65) μmol/mL] and SOD [(292.74±52.44), (303.57±13.80) U/mL] in serum and decrease the content of MDA [(15.60±2.39), (15.57±2.19) nmol/mL] significantly, and there were significant differences compared with model group (P 〈 0.05 or P 〈 0.01). Conclusion There is evident therapeutic effect of PSP on the experimental gastric ulcer in rats, and its mechanism may be related to scavenging oxygen free radical, improving the activity of SOD and decreasing the content of MDA.
