大黄酸对亚慢性镉染毒小鼠肝和肾损伤的保护作用

摘　　要：目的观察大黄酸对亚慢性镉染毒小鼠致肝、肾损伤的干预作用。方法 48只昆明种小鼠随机分为4组。单纯镉染毒模型组,大黄酸低剂量组,大黄酸高剂量组,正常对照组。除正常对照组外,3组实验小鼠皮下注射3.5mg/kg的氯化镉溶液染毒,每周3次,连续染毒4周,染毒的同时,大黄酸低剂量组每天给予大黄酸30mg/kg,大黄酸高剂量组每天给予大黄酸60mg/kg。正常对照组小鼠在相应时间皮下注射生理盐水。末次给药后处死小鼠,采血测定血清中丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)的活力。摘取小鼠肝脏和肾脏,测定肝和肾组织中丙二醛(MDA)的含量,超氧化物歧化酶(SOD)的活力。观察肝肾组织病理变化。结果与正常对照组比较,单纯镉染毒模型组肝、肾组织MDA含量显著升高,SOD活力显著下降。与单纯镉染毒模型组比较,大黄酸高剂量干预组肝、肾组织中的SOD活力显著升高,MDA含量降低,受损的肝、肾组织细胞的坏死明显减轻。结论大黄酸对亚慢性镉染毒小鼠所致肝、肾损伤有一定的保护作用,其机制可能与大黄酸抑制镉染毒小鼠体内氧化应激反应有关。Objective To study the effect of rhein on liver and renal damage induce by cadmium.Methods Totally 48 adult Kunming mice were randomly divided into four groups,control group,exposed cadmium chloride model group,high dosage rhein group,low dosage rhein group.Except control group was injected sc with 0.9% NaCl,other three groups were injected sc with3.5 mg/kg CdCl2,3 times a week,for up to 4 weeks.At the same time,high dosage rhein group administrated rhein 60 mg/kg and low dosage rhein group administrated rhein 30 mg/kg for 4 weeks.After the last administration,the levels of alanine aminotransferase（AST）,aspartate aminotransferase（ALT）,malondialdehyde（MDA）,superoxide dismutase（SOD）were determined.Results Compared with the control group,in exposed cadmium chloride model group,the content of MDA increased evidently,the activities of SOD obviously decreased.Compared with the exposed cadmium chloride model group,in the high dosage group,the content of MDA in liver and renal obviously decreased and the activities of SOD obviously increased.Conclusion Rhein has ability to prevent damage of liver and renal in mice that exposed in cadmium chloride and can inhibit the oxidative stress cause by cadmium chloride.
