摘 要:以润滑油为唯一碳源,从石油污染土壤中筛选分离得到了3株润滑油降解菌,根据形态特征和生理生化试验对菌株进行了鉴定,并考察了各分离菌株的润滑油降解性能。结果表明,所分离的3株菌株中,G1为黄单胞菌属(Xanthomonas),G2为动胶菌属(Azotobacter),G3为假单胞菌属(Pseudomonas);3株菌株均能生物降解润滑油,其中G3的润滑油降解能力最强。菌株G1、G2和G3降解润滑油的适宜pH值分别为8.0、7.0和8.0,适宜的降解温度分别为20、30和30℃。3株菌株适宜的接种量均为10%,且100mL培养液中润滑油的初始量不超过300μL。此外,培养时葡萄糖作为补充碳源,可不同程度地提高3株菌株的润滑油降解率。在最适降解条件,且1L液体培养基中添加2.0g葡萄糖时,菌株G1、G2和G3在3d内的润滑油降解率分别达到66.4%、75.3%和86.1%。With lube oil as sole carbon source, three lube oil biodegrading strains were screened from oil contaminated soil. The isolated strains were identified through morphological observations and physiological & biochemical tests, and their biodegradation characteristics were studied. The result showed that three isolated strains were Xanthomonas (named G1), Azotobacter (named G2) and Pseudomonas (named G3), respectively, which could biodegrade lube oil, and strain G3 had the highest biodegradation capability among the three strains. G1, G2 and G3 had the highest biodegradation capability at pH value 8.0, 7.0, 8.0 and at 20℃, 30℃, 30℃, respectively. Appropriate inoculation volume fraction for three strains was 10%, and appropriate initial lube oil volume in 100 mL liquid culture medium was less than 300 μL. Addition of glucose as co-carbon source variously accelerated lube oil degradation of three strains. When 2.0 g glucose was added to 1 L liquid culture medium, under the appropriate conditions the biodegradation capability of strain G1, G2, G3 within 3 d was 66.4%, 75.30% and 86. 1%, respectively.
