摘 要:通过优化色谱条件,建立了FOX-12纯度的高效液相色谱分析方法。采用C18反相色谱柱(5μm,250mm×4.6mm)分离样品,柱温为50℃,二元流动相乙腈-水体积比为60∶40,流速为0.8mL/min。结果表明,检测波长为280nm,在0.000 2~0.002g/mL范围内线性良好,线性相关系数为0.999 6,该法标准偏差为0.16,变异系数为0.68%,平均回收率为94.78%~97.53%,方法灵敏、准确,可用于FOX-12产品纯度分析。A high performance liquid chromatography (HPLC) method was established for determination of the purity of FOX-12 by optimizing the chromatographic conditions. The sample was separated by C18 column (5μm, 250mm×4.6mm) using binary mobile phase, acetonitrile and water (volume ratio of 60 : 40) with flow rate of 0.8mL/min at column temperature of 50℃. The results show that the UV detection wavelength is set at 280nm. A good linearity with correlation coefficient of 0. 9996 is obtained in the range of 0. 0002--0. 002 g/mL . The standard deviation of the method is 0.16, the coefficient of variation is 0.68%, and the average recoveries of the method are in the range of 94.78%- 97.53%, indicating that the method is sensitive and accurate, and can he applied in the determination of the purity of FOX-12.
