原发性与复发性骨肉瘤差异蛋白质组学研究

摘　　要：目的比较原发性与复发性骨肉瘤差异蛋白质,为寻找骨肉瘤复发的特异标记物提供依据。方法分别对5例骨肉瘤标本和4例复发性骨肉瘤提取总蛋白,然后双向凝胶电泳,重复3次,银染后扫描用Image2Master 2DE软件分析寻找差异蛋白,切除差异蛋白质点进行MS+MS/MS质谱鉴定,获取肽质指纹图谱并查询NCBI数据库寻找匹配蛋白质。结果获得重复性较好的双向电泳银染图谱,原发性与复发性骨肉瘤组织蛋白质点数分别为(1 206±161)和(1 387±185),找到明显差异的蛋白质点42个,并成功鉴定出13个。与原发性骨肉瘤相比,5个蛋白质在复发性骨肉瘤中表达下降;8个蛋白质在复发性骨肉瘤表达水平升高,其中β2微球蛋白在原发性骨肉瘤中表达缺失。结论原发性骨肉瘤与复发性骨肉瘤存在明显差异蛋白质,β2微球蛋白有可能成为诊断复发性骨肉瘤的分子标记物及检测指标。【Objective】 To compare the proteomics discrepancies between primary osteosarcoma and recurrent osteosarcoma for discovering specific markers for osteosarcoma.【Methods】 Total protein was extracted from five osteosarcoma samples and four recurrent sarcoma,respectively.Then dielectrophphoresis was conducted three times,repeatedly.To find out differential protein by Image 2 Master 2DE software analysis after silver staining and scanning.Then,MS ＋ MS / MS mass spectrometric identification of differential protein spots was conducted.Get access to the peptide mass fingerprints and query to find out matching proteins in the NCBIdatabase.【Results】 Dielectrophoresis silver staining map obtained had good repeatability.Protein spots of primary osteosarcoma and recurrent osteosarcoma are approximately（1 206 ± 161）,（1 387 ± 185）,respectively,and 42 significant different proteins were discovered,of which 13 spots were identified successfully.The recurrent osteosarcoma compare to the primary osteosarcoma,8 proteins were significantly up regulated,especially Macrophage migration inhibitory factor MIFand with the absence of β2-microglobulin（β2-MG） in the primary osteosarcoma;5 down-regulated proteins were observed.【Conclusion】 The results suggest that proteomic analysis can identify protein varianes between the primary osteosarcoma and recurrent osteosarcoma,β2-MG may serve as diagnostic markers and testing indicators.
