摘 要:目的:探讨COX-2和TGF-β1蛋白在乳腺癌变过程中的表达变化情况与意义。方法:采用免疫组化即用型二步法(非生物素)染色方法检测乳腺正常组织(61例)、乳管内乳头状瘤(65例)、原位癌(45例)及乳腺浸润性癌(87例)乳腺组织中COX-2、TGF-β1蛋白的表达情况。结果:总体表达情况上COX-2在乳头状瘤组织(6.26±2.56)、原位癌组织(7.23±3.02)及浸润性癌组织的表达(5.90±2.22)与正常乳腺上皮组织(4.73±2.03)相比,差异有统计学意义,P<0.01;乳头状瘤组织(2.15±0.97)与原位癌组织(2.52±1.18)的表达强度高于浸润性癌组织(1.68±0.86),P<0.01。TGF-β1总体表达情况在乳头状瘤组织(5.76±2.21)、原位癌组织(7.08±3.13)及浸润性癌组织的表达(6.77±2.54)与正常乳腺上皮组织的表达(4.88±2.13)相比,差异均有统计学意义,P<0.05;原位癌组织(2.23±1.16)及浸润性癌组织(2.21±1.09)表达强度高于乳管内乳头状瘤组织(2.01±0.91)与正常乳腺上皮组织(1.87±0.84),P<0.05。结论:COX-2及TGF-β1蛋白表达可能是乳腺癌癌变的早期事件,检测可能有助于临床的早期诊断及预防。OBJECTIVE: To detect the expression of COX-2,TGF-β1 protein with immunohistochemical method in normal breast tissues,intraductal papilloma (disease), carcinoma in situ, invasive cancer tissues, and to explore their ex- pression changes and significance in the development of breast cancer. METHODS: The immunohistochemistry (two step- method) was used to detect the expression of COX-2,TGF-~I protein in breast normal tissue group (n= 61),intraductal papilloma (disease) (n = 65 ), carcinoma in situ (n = 45 ), invasive breast cancer group (n = 87). RESULTS : The overall ex pression of COX-2 in papilloma (6.26±2.56) ,carcinoma in situ (7.23±3.02) ,invasive cancer tissues (5.90±2.22) was different statistically in compared with the normal breast epithelium(4.73±2.03 ,P〈0.05). The expression of COX-2 in papilloma (2.15± 0.97) and carcinoma in situ (2.52 ±1.18) was different statistically in compared with invasive cancer tissues (1.68± 0.86, P〈0.01). The overall expression level of TGF-β1 protein in papilloma(5.76 ± 2.21 ), carcinoma in situ (7.08±3. 13),invasive cancer tissues (6. 77±2.54) was different statistically compared with normal epithelium (4.88±2.13,P〈0.05). The expression intensity in carcinoma in situ (2.23±1.16) and invasive cancer tissues (2.21± 1.09) was higher than that in papilloma (2.01±0.91) and normal epithelium(1.87±0.84,P〉0.05). CONCLUSION: COX-2 and TGF-β1 protein expression may be an early event in breast cancer,and the detection of COX-2 may be contributed to early clinical diagnosis.
