摘  要:比较不同发育阶段猪苓菌丝、菌核显微结构和有效成分的含量。以卢氏县不同发育阶段的野生猪苓菌核为试验材料,用插片法和石蜡切片比较菌丝、菌核的显微结构;用超声提取和苯酚硫酸法测定菌丝菌核的多糖含量;用HPLC测定菌丝菌核的麦角甾醇含量。野生菌核随着生长年限的增加,菌丝的分化程度增高且结构致密,表皮木质化,同时多糖和麦角甾醇含量逐渐增高;蜜环菌的侵染诱导了菌核防御结构的发生,菌丝木质化并产生隔离腔,也有利于多糖和麦角甾醇的累积;液体发酵培养的菌丝体,其多糖和麦角甾醇的含量均高于各阶段的野生菌核。To compare hyphae and sclerotium microscopic structure,and the content of effective component of Polyporus umbellatus at different developmental stages,wild P.umbellatus sclerotium at different developmental stages collected from in Lushi County were used as test material.Microscopic structure of hyphae and sclerotium were compared by using insert method and paraffin section.Ultrasonic extraction and phenol-sulfuric acid method were used for determining the polysaccharide content of hyphae and sclerotium,while the ergosterol content was analyzed by HPLC.With the increase of the growth,higher degrees of differentiation of hyphae and dense structure were found,as well as epidermis lignification.Meanwhile,polysaccharides and ergosterol contents increased gradually.Additionally,the infection of Armillaria mellea induced the sclerotium of defensive structure,hyphae lignification and the formation of the isolation chamber.Besides,A.mellea infection was conducive to polysaccharides and ergosterol contents.Polysaccharide and ergosterol contents in hyphae cultured via liquid fermentation was found to be higher than those in wild sclerotium at each stage.
