摘  要:原生质体技术在遗传分析和育种中具有重要作用。金针菇双核体在制备原生质体过程中会出现单核化。为了解单核原生质体之间是否存在差异,对3个金针菇菌株单核体原生质体不同交配型菌株以及配对而成的双核体菌株进行了ISSR分析。结果表明,3个金针菇菌株2种不同交配型单核原生质体配对的双核体与出发菌株之间遗传关系较近,遗传相似系数高的为0.97、低的为0.75;3个金针菇菌株的不同交配型菌株之间遗传差异较大,分别为0.77、0.73和0.59,表明不同交配型的单核原生质体菌株之间遗传差异显示其遗传背景的不同。Protoplast technique plays an important role in genetic analysis and breeding.Monokaryogenesis of Flammulina velutipes dikaryons probably appeared in theirs protoplast preparation.3 mononuclear strains of Flammulina velutipes of different mating types and their corresponding dikaryons strains were analyzed by ISSR to reveal the potential differences among mononuclear protoplasts.The result showed that the genetic relationships between dikaryons and their original strains were relatively close with genetic similarity coefficients between 0.75 and 0.97;the protoplasted mononuclear strains of 3 different mating types had considerable genetic diversity,and their genetic similarity coefficients were 0.77,0.73 and 0.59,respectively,which indicated their different genetic backgrounds.
