摘 要:目的研究抑癌基因WT-1在子宫颈癌组织中的表达及意义。方法子宫颈癌患者80例(转移癌40例、未转移癌40例),正常宫颈组织40例,以GAPDH作为内参基因,采用Taqman探针实时荧光定量逆转录聚合酶链反应(RT-PCR)技术检测转移抑制基因WT-1在各组中的表达情况,结合临床、病理资料分析其与宫颈癌发生发展的相关性。结果抑癌基因WT-1在正常子宫颈组织中的表达量(0.0183±0.0074)高于子宫颈癌未转移组(0.0035±0.0015)、子宫颈癌转移组(0.0030±0.0012),差异均有统计学意义(P均<0.01),后两者比较差异无统计学意义(P>0.05)。WT-1基因在转移组Ⅰa、Ⅰb、Ⅱa期中的表达分别为(0.0030±0.0002)、(0.0029±0.0001)和(0.0029±0.0004),在未转移组Ⅰa、Ⅰb、Ⅱa期中的表达分别为(0.0037±0.0016)、(0.0031±0.0016)和(0.0034±0.0003)两两比较差异均无统计学意义(P>0.05)。结论抑癌基因WT-1参与子宫颈癌的发生过程,但与子宫颈癌转移的关系不明显。Objective To study the expression condition and significance of tumor suppressor gene WT-1 in cervical cancer ceils. Methods With GAPDH as reference, using Taqman probe real time quantitative reverse transcription polymerase chain reaction detects the expression level of WT-1 gene in 80 cases cervical cancer tissues, in which 40 cases with metastasizing and 40 cases without metastasizing, and 40 cases normal cervical tissues, analysis the relationship between the occurring of cervical cancer and WT-1 gene expression with clinical dates and pathologic dates. Results WT-1 expression level in normal (0.0183 ±0. 0074) was more than non-metastasizing (0. 0035 ±0.0015), difference was significant ( P 〈0.01 ) ; WT-1 expression level in normal was more than metastasizing (0. 0030 ±0.0012), difference was significant ( P 〈0.01 ) ; the difference of WT-1 expression level between non metastasizing and metastasizing has no significant ( P 〉 0.05). WT-1 gene transfer group Ⅰa, Ⅰb, Ⅱa expression period were (0.0030 ± 0. 0002 ), (0. 0029± 0. 0001 ) and (0. 0029 ±0. 0004 ), without metastasis group Ⅰa, Ⅰb, Ⅱa expression period were (0. 0037 ± 0. 0016), (0. 0031 ± 0. 0016) and (0. 0034 ± 0. 0003), pairwise comparisons showed no significant difference ( P 〉0.05). Conclusion Tumor suppressor genes WT-1 would participate in the process of the occurrence of cervical cancer, but the relationship with cervical cancer metastasis is not obvious.
