摘 要:目的探讨细胞中富亮氨酸糖蛋白(LRG)高表达对转化生长因子β1(TGF-β1)、转化生长因子受体体Ⅱ(TβR-II)的影响。方法设计构建pAcGFP1-C1-LRG质粒,设为转染组和对照组,转染组将pAcGFP1-C1-LRG质粒转染入U87脑胶质瘤细胞,分别于转染后48 h及72 h收集细胞,提取总RNA,应用Real-time PCR方法测定U87细胞中LRG、TGF-β1及TβR-Ⅱ的mRNA表达。结果 pAcGFP1-C1-LRG质粒转染入U87细胞后LRG表达明显增高,TGF-β1、TβR-II的表达明显高于对照组,且转染后72 h表达高于48 h,差异均有统计学意义(P<0.05)。结论 LRG在细胞内高表达能促进TGF-β1、TβR-II的表达。Objective To study the effect that the overexpression of leucine rich α2 glycoprotein (LRG) on the TGF-β1 and TβR-Ⅱ. Methods Designed and structured the pAcGFP1-C1-LRG plasmids, then the plasmids were transfected into human U87 glioma cells. After transfection 48 hours and 72 hours, we respectively collected cells, and extracted the total RNA In order to analyze the expression of LRG,TGF-β1 and TβR-Ⅱ in the U87 glioma ceils, the real time PCR was performed. Results After the plasmids were transfected into human U87 glioma cells, all expressions of LRG, TGF-β1 and TβR-Ⅱ were increased significantly, compared with the control group. Conclusion These results suggest that the overexpression of LRG in cells may promote the expression of TGF-β1 and TβR-Ⅱ.
