摘 要:目的:研究维持性血液透析患者乙型肝炎病毒(HBV)感染基因型的分布,对HBV-DNA S基因进行同源性分析。方法:聚合酶链式反应(PCR)对乙肝表面抗原(HBsAg)阳性者进行HBV-DNA S基因扩增,双脱氧末端终止法测序,MEGA5.1软件构建系统发生树行基因型及同源性的分析。结果:HBsAg阳性率20.8%;24例样本PCR扩增出阳性产物,HBV基因型B型13例,C型11例;系统进化分析提示,2例HBV-DNA S基因进化距离0.000,相似度100%。结论:维持性血液透析者HBsAg阳性率较高,存在B、C型两种HBV基因型,2例HBV-DNA S基因同源性极高,提示为同一病毒株感染。Objective: To study the distribution of genotypes of hepatitis B virus (HBV) from maintenance hemodialysis patients, and to analyze homology of HB V-DN.4 S gene. Methods: HBV-DNA S gene was obtained by polymerase chain reaction(PCR) amplifying and sequencing by dideoxy termination method in maintenance hemodialysis patients with HBsAg positive. The gene sequences were analyzed and the polygenetic tree was drawn by the software MEGA 5.1. Results: Frequency of HBsAg positive was 20.8%. Thirteen cases were identified as genotype B and the remaining 11 cases were genotype C in the 24 S gene PCR positive serums. Polygenetic analysis revealed that the evolutionary distance was 0.000, similarity 100% between HBV-DNA S gene of two patients. Conclusion: HBsAg positive rat is high, and both genotype B and C are prevailed in the dialysis unit. HBV-DNA S gene are highly homologous in two patients, which suggests that the same strain of the virus infection.
