芳香开窍法对大鼠脑缺血再灌注后血脑屏障超微结构损伤及AQP-4表达的影响

摘　　要：目的通过观察大鼠脑缺血再灌注后血脑屏障(BBB)超微结构损伤及AQP-4表达情况,探讨芳香开窍法对BBB的保护作用。方法建立大鼠局灶性脑缺血再灌注模型,电镜下观察BBB超微结构,分别用RT-PCR方法、免疫组化方法研究AQP-4 mRNA、AQP-4蛋白表达的变化。结果与模型组比较,麝香配伍冰片组AQP-4 mRNA表达量显著降低(P<0.01),冰片组AQP-4 mRNA表达量也较模型组下降(P<0.05)。麝香配伍冰片组AQP-4 mRNA表达量较麝香组低,差异有统计学意义(P<0.05)。麝香配伍冰片组AQP-4蛋白表达量较模型组下降明显,差异有统计学意义(P<0.05),冰片组AQP-4蛋白表达量也较模型组下降(P<0.05)。结论麝香配伍冰片可减轻缺血再灌注后BBB超微结构损伤,抑制脑组织AQP-4 mRNA和蛋白的表达而起到脑保护作用,这可能是其芳香开窍的作用原理之一。Objective To discuss the protection effect of resuscitation-inducing therapy with fragrant drugs on Blood Brain Barrier （BBB） by ultrastructure observation and investigation of AQP-4 expression in rats with isch- emia/reperfusion. Methods Focal cerebral ischemia/reperfusion model （MCAO） rat was set up first of all, then the obser- vation ofBBB ultrastmcture using electron microscopy was done. The expression of AQP-4 mRNA was measured utiliz- ing real-time PCR （RT-PCR） and its protein expression was evaluated utilizing immunohistochemistry. Results Musk with bomeul could effectively reduce BBB endothelial cell swelling, loose fight junctions, basement membrane struc- ture and glial cell line-fuzzy edema after ischemia/reperfusion. AQP-4 mR.NA expression in musk with bomeul group, borneo1 group were significantly inhibited, AQP-4 protein expression in musk with bomeol group, bomeol group de- creased significantly compared with model group, Conclusion Musk with Bomeol could reduce BBB damage and its permeability and brain edema through inhibiting the expression ofAQP-4 mRNA and protein after ischemia/reper- fusion. That may be one of its mechanisms of resuscitation-inducing therapy with fragrant drugs.
