液氮冻存不同时间大鼠带瓣管道对其活性和组织中IGF-1水平的影响

摘  要:目的:观察、研究液氮冻存不同时间的大鼠带瓣管道的活性和其组织中胰岛素样生长因子(IGF-1)的水平,提供一种检测带瓣管道活性的新思路。方法:Wistar大鼠60只,随机分为新鲜组(A组),-196℃液氮冻存1个月(B组),冻存3个月(C组)。取材处理后检测其葡萄糖代谢、管道结构及组织中IGF-1水平的变化。结果:大鼠带瓣管道的3组葡萄糖代谢率测定,A组(3.812±0.476 8)mmol/L、B组(3.826±0.349 5)mmol/L、C组(3.655±0.373 6)mmol/L比较,差异无统计学意义(P>0.05);IGF-1在带瓣管道中的表达分别为35%、30%、30%,差异无统计学意义(P>0.05)。结论:液氮冻存后的大鼠带瓣管道活性良好,提示IGF-1的表达水平可作为评价带瓣管道活性的指标。Objective:To investigate the effects of rat valved conduits after different time cryopreservation on their activity and tissue IGF-1 content, and attempt a new test method of valved conduit activity. Meth- ods. Sixty Wistar rats were randomly divided into three groups. Twenty cases of fresh blood vascular (group A), 20 cases of frozen vessels for 1 month (group B), 20 for 3 months (group C). The changes of glucose metabolism rate and the IGF-I content of the vascular were investigated. Results. The glucose me- tabolism rate of valve conduit in groups A,B and C was(3. 812±0. 476)mmol/L, (3. 826±0. 349)mmol/L, and (3. 655±0. 373)mmol/L ( P 〉0.05), respectively. The positive expression rate of IGF-I in groups A, B and C was 35%, 30% and 30% ( P 〉0.05), respectively. Conclusion: The rat valved conduits have good activity after frozen at different time points, and IGF-I is one of the indices of valved conduit activity.
