摘 要:目的观察经TGFβ1基因修饰的人骨髓间充质干细胞(hMSC)对高转移潜能肝癌细胞(MHCC97-H)增殖能力及侵袭能力的影响。方法构建TGFβ1慢病毒载体,eGFP为报告基因,转染hMSC。Transwell法检测hMSC与肝癌细胞共培养实验,检测MHCC97-H细胞侵袭能力的变化。CCK-8法检测与hMSC共培养前后MHCC97-H增殖能力的变化。PCR法检测转基因hMSC与MHCC97-H细胞共培养前后转移相关因子基因α-V、肿瘤生长因子TGFβ1和肿瘤凋亡因子PDCD4等基因表达。结果 MHCC97-H与hMSC细胞共培养后,MHCC97-H增殖能力增强,hMSC TGFβ1基因过表达组对MHCC97-H细胞增殖具有抑制作用。MHCC97-H与hMSC细胞共培养后,侵袭能力增强,hMSC TGFβ1基因过表达组对MHCC97-H细胞侵袭能力具有明显的抑制作用。hMSC基因转染组TGFβ1表达明显增高。hMSC基因转染共培养组MHCC97-H细胞TGFβ1基因表达明显低于对照组。hMSC基因转染共培养组MHCC97-H细胞α5基因表达降低。hMSC基因转染共培养组MHCC97-H细胞PDCD4基因表达降低。结论转基因hMSC具有抑制MHCC97-H细胞增殖、侵袭的作用。Objective To observe the TGFβ1 gene-modified human bone marrow mesenchymal stem cells (hMSCs) impacting on high metastatic HCC ceils (MHCC97-H) proliferation and invasion ability. Methods TGFβ1 lentiviral vector was structured, eGFP as reporter gene transfected hMSC. MHCC97-H cell invasion ability of hMSC with MH- CC97-H cells co-culture experiments was detected by Transwell assay. Proliferative capacity of MHCC97-H co-culture test was detected by CCK-8 assay. The relevant factor gene alpha-V, tumor growth factor TGFβ1 and tumor apoptotic factors PDCD4 gene expression of transgenic hMSC with MHCC97-H ceils co-culture were detected by PCR assay. Results Proliferation ability was increased after MHCC97-H co-cultured with hMSC cell. TGFβ1 gene overexpression of hMSC group inhibited cell proliferation of MHCC97-H. Invasion ability after MHCC97-H co-cultured with hMSC cells was increased, hMSC of TGFβ1 overexpression group significantly inhibited cell invasion ability of MHCC97-H. TGFβ1 gene expression of MHCC97-H cells with hMSC gene transfection co-culture group was significantly lower than that in the control group, or5 gene expression of MHCC97-H with hMSC gene transfected cells co-cultured group was decreased. PDCD4 gene expression of MHCC97-H with hMSC gene transfected ceils co-cultured group was reduced. Conclusion Genetically modified hMSC have inhibitory action of the MHCC97-H cell proliferation and invasion.
