探讨Peroxiredoxin1在胃癌腹膜转移中的作用机制

摘　　要：目的探讨胃癌组织中Peroxiredoxin1(Prx1)的表达与胃癌腹膜转移的关系。方法采用免疫组织化学SP染色法检测上海交通大学附属新华医院收治的65例同期伴腹膜转移胃癌患者的胃癌原发灶、腹膜转移灶、淋巴结转移灶及癌旁正常胃黏膜组织中prx1蛋白的表达。结果免疫组化显示胃癌原发灶、腹膜转移灶及淋巴结转移灶中prx1蛋白强表达,阳性表达率分别为胃癌原发灶60%(39/65)、腹膜转移灶68%(44/65)、淋巴结转移灶88.6%(39/44),癌旁正常胃黏膜组织中Prx1蛋白阳性表达率为3%(2/65),且表达很弱。癌旁正常胃黏膜中prx1蛋白的表达与胃癌原发灶、腹膜转移灶及淋巴结转移灶之间比较,差异具统计学意义(P<0.05);胃癌原发灶、腹膜转移灶及淋巴结转移灶之间比较,差异无统计学意义(P>0.05)。结论胃癌病灶中的Prx1检查可能成为判断胃癌术前是否腹膜转移以及预后的生物学标志。Objective To investigate the relationship between the expression of Peroxiredoxinl（Prxl） and peritoneal metastasis of gastric cancer. Methods sixty-five patients who had gastric cancer and with peritoneal metastasis had been admitted to Shanghai jiaotong university of medicine Xinhua Hospital. Gastric cancer tissues,metastatic peritoneal tissues, metastatic lymph nodes and adjacent tissues were obtained postoperatively, and the expression of Prxl was detected by immunohistochemistry. The relationship between Prxl expression and clinicopathological characteristics of gastric cancer was analyzed. Results The expression of Prxl Was mainly detected in the cell cytoplasm. The positive expression rates of Prxl in gastric cancer tissues, metastatic peritoneal tissues and metastatic lymph nodes were 60%（39/65）,68%（44/65）and 88.6%（39/44）,respectively, whereas in 3%（2/65）of paratumor normal tissues. There were significant differences in the Prxl expression between adjacent tissues and gastric cancer tissues, metastatic peritoneal tissues ,metastatic lymph nodes respectively.（P〈0.05）,but there were no differences among gastric cancer tissues, metastatic peritoneal tissues and metastatic lymph nodes respectively.（P〉0.05）.Conclusions The expression of Prx 1 in gastric cancer lesions may be used as a biological marker of peritoneal metastasis from gastric cancer before operation and as a prognostic factor cancer.
