摘 要:目的观察沙利度胺对小鼠T淋巴瘤的抑制作用。方法 32只BALB/C小鼠皮下接种鼠T淋巴瘤白血病细胞(EL-4细胞)建立模型,第2天随机分成4组,每组8只:①阴性对照组:小鼠8只,生理盐水灌胃25 mL/(kg.d);②低剂量组:小鼠8只,沙利度胺灌胃50 mg/(kg.d)共14 d;③中剂量组:小鼠8只,沙利度胺灌胃200 mg/(kg.d)共14 d;④高剂量组:小鼠8只,沙利度胺灌胃400 mg/(kg.d)共14 d。每2日测肿瘤直径,计算体积,用药后15 d处死小鼠,分离肿瘤,计算抑瘤率。第2批小鼠同样方法处理,记录平均生存期。用TUNEL法检测肿瘤细胞凋亡指数。RT-PCR法测TNF-αmRNA表达。结果各组瘤重较阴性对照组减小,抑瘤率分别为17.75%,36.73%,42.04%,差异有统计学意义(P<0.05)。各组生存期与阴性对照组相比延长,差异有统计学意义(P<0.05)。沙利度胺各组肿瘤坏死因子表达下调,凋亡细胞增多,差异有统计学意义(P<0.05)。结论沙利度胺能抑制小鼠T淋巴瘤生长,延长荷瘤小鼠平均生存期,而抑制TNF-α的表达,诱导凋亡可能是其机制之一。Objective To observe the effect of Thalidomide on T-lymphoma in mice. Methods The mouse lymphoma model was established by injecting EL--4 cells subcutaneous in 32 BALB/C mice. Then the mice were divided randomly into four groups on the second day after the injection, every group had 8 mice. (1)eontml group: NS 25 mg](kg.d), ig;, (2)low dose group: Thalidomide 50 mg/(kg'd), ig for 14 days; (2)middle dose group: Thalidomide 200 mg/(kg.d), ig for 14 days; (1)high dose group: Thalidomide 400 mg/(kg 'd), ig for 14 days; the diameters of the tumors were measured every two days. On the 15th day, the mice were sacrificed and the tumors were weighed, and tumor inhibition rate was calculated. The survival time of the tumor-bearing mice were record on another same method. The index of apoptosis (AI) of tumor tissue was evaluated by the method of terminal deoxynucleotidyl transferase biotin-dUTP nick end label- ing (TUNEL). Expression of TNF-a mRNA was tested by reverse transcription-polymerase chain reaction (RT-PCR). Results The tumor weight in Thalidomide treatment groups decreased compared with the control group, the difference had statistical significance (P 〈 0.05). The inhibition rates were 17.75%, 36.73%, 42.04% respectively in Thalidomide- treated groups. The survival time of the tumor-bearing mice in Thalidomide-treated groups was significantly longer than that of control group (P 〈 0.05). Compared with the negative control group, the relative expression of TNF-a mRNA of the Thalidomide-treated groups significantly decreased. The index of apoptosis (AI) of the Thalidomide treatment groups were significantly higher (P 〈 0.05). Conclusion Thalidomide can inhibit T-lymphoma growth and prolong the survival time in mouse model. One of the mechanisms is probably that Thalidomide inhibite the expression of TNF-a and increase the index of apoptosis.
