摘  要:旨在研究总丹参酮包合物的体外抑菌作用。采用乙醇回流方法提取并以大孔吸附树脂进行分离纯化,HPLC测定含量,以β环糊精制备成包合物。选用大肠杆菌、金黄色葡萄球菌等几种常见致病菌作为试验菌。平板菌落计数法筛选1×108 cfu/mL菌液涂板,以杯碟法测定体外抑菌活性,琼脂稀释法测定最小抑菌质量浓度(MIC)。结果表明:总丹参酮包合物对各试验菌株均有不同程度的抑制效果。对金黄色葡萄球菌、枯草芽孢杆菌和停乳链球菌的抑制效果显著。总酮包合物中,丹参酮ⅡA和隐丹酮对金黄色葡萄球菌的MIC值分别为0.73和2.51mg/L;二者对枯草芽孢杆菌和停乳链球菌的MIC值相同,丹参酮ⅡA为1.18mg/L,隐丹参酮为4.02mg/L。总丹参酮包合物具有较强的抗菌作用。同时,总丹参酮包合物具有良好的溶解性,改善丹参总酮提取物难溶于水、吸收差的特性,提高其生物利用度,可进行生物制剂研究及配合临床抗生素使用,具有较高的应用前景。This research aimed to study the in vitro antimicrobial activity of the total tanshinones-β-cyclodextrin. After using the refluent extraction with ethanol, extracted total tanshinones was separated and purified with macroporous resin. The content of total tanshinones was determined by HPLC.β-cyclodextrin is used as inclusion material to obtain the total tanshinones-β-cyclodextrin. Several types of common pathogenic bacteria were selected as the tested strains to evaluate the inhibition effects of total tanshinones-β-cyclodextrin. The bacteria liquid with 1x 108 cfu/mL which was prepared by fiat calculating colony was smeared on the MH bacteria in order to evaluate the inhibitory rate and minimum inhibitory mass concentration(MIC) by using the cylinder plate method and agar dilution method. It was found that the total tanshinones-β-cyclodextrin was actively against all types of tested pathogenic bacteria and exhibited a high sensitivity to Staphyloccocus aureus ,Bacillus subtilis and Streptococcus dysgalactiae subsp equisimilis. In total tanshinones-β-cyclodextrins, the MIC of tanshinone Ⅱ A and Cryptotanshinone to Staphylococcus aureus are 0.73 mg/L and 2.51 mg/L. To Streptococcus dysgalactiae subsp equisimilis and Bacillus subtilis, tanshinone Ⅲ A and Cryptotanshinone have the same MIC of 1.18 mg/L and 4.02 mg/L. The total tanshinones-β-cyclodextrin had strong antibacterial action on many types of bacterium. At the same time, it had good solubility and had changed the poorly water-soluble character of total tanshinones, improved the weak absorption of total tanshinonesd and raised the level of biologic utilization. This study can provide significant basis for developing some biological agents and may instruct the application of the antibiotics reasonably in clinical use. It had brilliant prospects of utility and was worth for further study.
