摘  要:以暗培养7 d的蓖麻上胚轴为外植体,通过研究不同浓度TDZ对上胚轴不定芽诱导的影响,以及BA和GA3不同配比对不定芽伸长的影响,建立高效的蓖麻体外繁殖体系。芽诱导阶段,上胚轴接种在添加了TDZ的MS(Murashige and Skoog)培养基上,结果表明,TDZ质量浓度为0.14 mg/L时,诱导的不定芽个数最多(24.07),同时出芽率也较好(80.61%);TDZ质量浓度为0.11 mg/L时,不定芽个数为20.06,出芽率最高(82.38%)。根据诱导试验表明,添加活性炭(AC)和0.30 mg/L的1/2 MS培养基,根诱导效果最好,达95%以上。移栽后,植株成活率为80%。In this present study, the effect of all concentrations of TDZ (Thidiazuron) on shoot induction and the effect of combination of BA and GA3 on shoot elongation are studied. As a result, an efficient protocol of castor tissue culture with epicotyl explant is established. Epicotyl is incubated on MS (Murashige and Skoog) medium. For shoot induction, 0.14 mg/L TDZ gives the maximum number of shoots (24.07) with the second best frequency of explants producing shoots (80.61%) ; while 0.11 mg/L TDZ with a high number of shoots (20.06) is found best for the frequency of explants producing shoots (82.38%) . Strong healthy shoots ere incubated on half strength of MS medium supplemented with 0.30 mg/L IBA and 1% AC with 95% root induction rate. About 80% of the plantlets ere survived after transplanting into soil.
