摘  要:目的研究5-脂氧合酶激活蛋白(ALOX5AP)基因SG13S114位点多态性与吸烟在中国汉族人群缺血性脑卒中发病中的交互作用。方法采用聚合酶链反应限制性内切酶长度多态性(PCR-RFLP)技术及非条件Logistic回归模型,分析基因型、吸烟情况与缺血性脑卒中的关系。结果病例组AT+AA基因型频率高于对照组,差异有显著性(χ2=5.986,P<0.05)。AA和AT基因型增加了缺血性脑卒中的发病风险(OR=1.994、1.391)。在调整混杂因素后,单独携带AA+AT基因型者OR值为1.663(95%CI=1.048~2.640),单独吸烟暴露者OR值为2.672(95%CI=1.630~4.382),吸烟和携带AA+AT基因型同时存在者OR值为5.050(95%CI=3.031~8.414),其交互作用系数为1.647。结论 ALOX5A基因SG13S114A/T多态性与吸烟的交互作用增加了缺血性脑卒中的发病风险。Objective To investigate the interaction between ALOX5AP SG13Sl14 gene polymorphism and smoking on ischemic stroke (IS) in Chinese Han population. Methods The relationship between genotype and cigarette smoking and ische- mic stroke was analyzed by using PCR-RFLP and unconditional logistic regression. Results The frequency of AT+AA genotype in IS group was significantly higher than that in the controls (x2 =5.986,P〈0.05); the AA and AT genotypes increased the risk of IS (OR=1.994,1.391). After adjustment of confounding factors, the OR value in those with AT--AA genotypes alone was 1.633 (95%CI=1.048--2.640) ; that in smoking only was 2.672 (95%CI= 1.630--4.382) ; that in both smoking and AA+AT genotypes was 5.050 (95 %CI=3.031--8.414)with interaction coefficient being 1.647. Conclusion The interactions between ge netic polymorphism of ALOX5AP SG13S114A/T and smoking increase the onset risk of ischemic stroke.
