卡介苗对大鼠膀胱肿瘤相关细胞因子表达的影响

摘　　要：目的通过检测卡介苗(BCG)作用大鼠膀胱肿瘤细胞后细胞因子的变化情况,探讨卡介苗(BCG)对膀胱肿瘤细胞的抑制作用。方法构建大鼠膀胱肿瘤模型,并原代培养大鼠膀胱肿瘤细胞。分别用普通培养液、卡介苗(BCG)作用于大鼠膀胱肿瘤细胞,ELISA法检测两组细胞上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)、干扰素-γ(IFN-γ)的水平。结果两组细胞上清液中IL-10、TNF-α水平比较,差异有显著性(t′=7.51、78.24,P<0.05)。而两组细胞上清液中IFN-γ水平比较,差异无显著性(P>0.05)。结论 BCG可以直接刺激肿瘤细胞分泌细胞因子,并对肿瘤细胞具有抑制作用。Objective To explore the effect of Bacillus Calmette Guerin （BCG） on inhibition of bladder cancer cells through detecting the level of cytokine after application of BCG to bladder tumor in rats. Methods A bladder cancer model was created in rats, the cancer cells were primarily cultured. Ordinary culture solution and BCG were respectively used to act on the cells. The concentrations of IFN-γ, TNF-α, and IL-10 in the cell supernatant in both groups were detected with ELISA. Results The differences of IL -10 and TNF-α level between the two groups were significant （t＇=7.51,78.24;P〈0.05）, but that of IFN-γ was not （P〉0.05）. Conclusion BCG can directly excite cancer cells to secrete cytokine and inhibit the cells.
