摘 要:目的探讨胺碘酮对兔心肌缺血再灌注后血清超氧化物歧化酶(SOD)和丙二醛(MDA)水平的影响及意义。方法将新西兰大白兔随机分为假手术组、对照组、胺碘酮组,假手术组开胸只穿线不结扎血管,后两组开胸并结扎兔左冠状动脉回旋支制备缺血再灌注无复流模型,每组10只。对照组再灌注即刻给予等量的生理盐水,胺碘酮组再灌注即刻给予胺碘酮3mg/kg。分别于再灌注后120min取血,测定各组血清SOD活性和MDA含量。结果在缺血范围差异无显著性的条件下,胺碘酮组无复流范围显著小于对照组(t=4.483,P<0.05)。与假手术组相比,对照组及胺碘酮组再灌注后血清中SOD活性明显降低(F=800.90,q=18.89、55.65,P<0.01),MDA含量明显增加(F=1 457.60,q=55.13、73.32,P<0.01);与对照组相比,再灌注后胺碘酮组血清中SOD活性明显增高(q=36.76,P<0.01),MDA含量明显降低(q=18.18,P<0.01)。结论胺碘酮可能是通过抑制氧自由基的产生,改善心肌缺血再灌注损伤,减轻无复流损伤的程度。Objective To assess the influence of Amiodarone on serum superoxide dismutase (SOD) and malondialde hyde (MDA) in rabbits and its significance after myocardial ischemia/reperfusion (I/R). Methods Thirty mature rabbits were evenly randomized to sham-operation group, control group and amiodarone group. Open chest surgery onl^without legating the blood vessels-was done for rabbits in the sham-operation group. For those in the latter two groups, left circumflex arteries were li- gated to create an I/R model. The model in the control was given reperfusion of equivalent normal saline, and those in amiodarone group were offered amiodarone 3 mg/kg. Serum SOD and MDA were measured after 120min of reperfusion. Results Under si- milar condition of ischemic range, the no-reflow area in amiodarone group was much smaller than that in the control (t = 4.483, P〈0.05). Compared with shame operation group, the activity of serum SOD in the control and amiodarone groups after re-perfu- sion was significantly lowered (F:800.90;q=18.89,55.65;P〈0.01), and MDA elevated (F:I 457.60;q--55.13,73.32;P〈0.01) ; Compared with the control, the activity of SOD in amiodarone group was significantly elevated (q- 36.76, P 〈0.01 ), and content of MDA lowered (q=18.18, P 〈 0.01). Conclusion Amiodarone relieves the extent of no-reflow damage is probably through inhibiting the generation of oxygen-derived free radicals and ameliorating myocardial I/R injury.
