摘 要:目的探讨非小细胞肺癌血清中白细胞介素8(interleukin-8,IL-8)和Fractalkine表达在肿瘤侵袭转移过程中的作用机制。方法选取非小细胞肺癌60例,其中伴淋巴结转移30例,无淋巴结转移30例,同时选取正常肺组织30例,采用双抗体夹心ELISA法测定其血清中趋化因子IL-8和Fractalkine的表达,对比分析与临床病理参数的关系。结果①IL-8和Fractalkine在非小细胞肺癌患者血清中的含量明显高于正常组,差异有统计学意义(P<0.01);IL-8和Fractalkine在淋巴结转移肺癌患者血清中的含量明显高于无淋巴结转移组,差异有统计学意义(P<0.05)。②IL-8和Fractalkine在非小细胞肺癌患者血清中的含量间存在正相关性(r=0.680 2,P<0.05)。③肺癌组IL-8的血清含量与肿瘤淋巴结转移、脉管内癌栓、临床TNM分期密切相关(P<0.05)。④肺癌组Fractalkine的血清含量与肿瘤淋巴结转移、脉管内癌栓、浸润肺膜密切相关(P<0.05)。结论 IL-8和Fractalkine的表达与肺癌的发展和远处转移密切相关,并对淋巴结转移发挥协同作用;血清学检测可预测肿瘤的转移潜能。Objective To investigate the effect of serum interleukin-8 (IL-8) and Fractalkine in the process of invasion and metastasis of non small cell lung cancer. Methods Sixty patients with non- small cell lung cancer, including 30 cases without node metastasis and 30 cases without lymph node metastasis respectively. The expression of serum IL-8 and Fractalkine were examined by double antibody sandwich ELISA method. The correlation between IL-8 and Fractalkine expression and clinical pathological parameters were analyzed. Results (1)IL-8 and Fractalkine of patients with non-small cell lung cancer were significantly higher than these of normal group, the differences were significant (P 〈 0.01 ) ; IL-8 and Fractalkine of lymph node metastasis patients were significantly higher than those of patients without lymph node metastasis, the difference were significant ( P 〈 0. 05 ). (2) There was a positive correlation between IL-8 and Fractalkine of patients with non-small cell lung cancer ( r = 0.680,P 〈 0. 05 ). (3)In lung cancer group, IL-8 level was closely related to tumor lymph node metastasis, intravascular cancer embolus, and clinical TNM staging ( P 〈 0.05 ). (4)In lung cancer group, Fractalkine level was closely related to tumor lymph node metastasis, intravascular cancer embolus, and pulmonary infiltration membrane ( P 〈 0.05 ). Conclusion The expression of IL-8 and Fractalkine are closely related with the development and metastasis of lung cancer, and exert synergistic effect on lymph node metastasis. The serol9gical test can predict the metastasis Dotentialitv of tumor.
