摘 要:目的探讨人胃癌SGC7901细胞中干细胞对顺铂耐药的作用机制。方法胃癌顺铂耐药细胞株SGC7901/DDP为实验组,胃癌SGC7901细胞为对照组,分别以Hoechst33342染色法、免疫组织化学、Tran-swell小室检测实验组及对照组的SP细胞(side population,SP)比例、Nanog的表达及细胞侵袭力。结果实验组中细胞株侧群SP细胞的比例为(5.51±1.68)%,显著高于对照组(2.45±0.63)%(P<0.05);实验组中Nanog的阳性表达率为(79.06±10.37)%,显著高于对照组(55.18+3.64)%(P<0.05);实验组穿膜细胞数为(65±7)个,显著高于对照组(41±6)个(P<0.05)。实验组胃癌细胞的生长抑制率均显著低于对照组(P<0.05);顺铂对实验组和对照组细胞的IC50值分别为(3.9±1.1)μmol/L和(25.7±2.3)μmol/L,实验组显著高于对照组(P<0.05)。结论胃癌干细胞可能与胃癌对顺铂耐药的机制密切相关,其中胃癌干细胞标志物SP细胞比例增高、Nanog基因高表达及细胞侵袭力增强发挥着重要作用。【Objective】 To investigate the mechanism on Cisplatin resistance of stem cell in human gastric cancer SGC7901 cell.【Methods】 Gastric cancer Cisplatin resistant cell line SGC7901/DDP was established to be as experimental group,and SGC7901 cell was as control group.The percentage of SP cells in SGC7091 cell line was detected by Hoechst 33342 staining.Immunocytochemical staining was used to detect the expression of Nanog.Invasion effect was detected by Transwell-system.【Results】 The ratio of SP cells in experimental group was(5.51 ± 1.68).It was significant higher than that of control group(2.45 ± 0.63) %(P &lt;0.05).The expression of Nanog in experimental group and control group were(79.06 ± 10.37) % and(55.18 + 3.64) % respectively.There had a significant difference between the two groups(P &lt;0.05).The number of cells passed through film in experimental group and control group were(65 ± 7) and(41 ± 6) respectively.Compared with the control group,the number of cells passed through film in experimental group was significantly higher(P &lt;0.05).The growth inhibition rate of gastric cancer in experimental group was significantly higher than that of control group(P &lt;0.05).IC50 value of experimental group and control group were(3.9 ± 1.1) μmol/L and(25.7 ± 2.3) μmol/L respectively.【Conclusion】 The mechanism on Cisplatin resistant may be associated with those gastric cancer stem cell markers,including increased proportion of SP cell,high expression of Nanog gene and enhanced cell invasion.
