摘 要:目的探讨线粒体DNA的ND-1基因G3316A和T3394C突变与老年2型糖尿病的相关性。方法采用PCR-基因测序技术,对96例老年T2DM患者(糖尿病组)和100例糖耐量正常的健康老人(正常对照组)进行检测。结果糖尿病组中检出G3316A突变4例(4.17%),正常对照组检出1名(1.04%),两组间比较差异有统计学意义(P<0.05)。糖尿病组中检出T3394C突变3例(3.13%),正常对照组检出1名(1.04%),两组间比较差异有统计学意义(P<0.05)。发生G3316A突变的患者与发生T3394C突变的患者之间无交叉突变。结论线粒体DNA的ND-1基因G3316A和T3394C突变与老年人T2DM的发生有关。Objective To explore the correlation between G3316A,T3394C mutation in the mitochondrial DNA ND-1 gene and type 2 diabetes mellitus(T2DM) in elderly patients. Methods PCR-gene sequencing technology was used to detect G3316A,T3394C mutation in 96 elderly patients with T2DM(diabetic group) and 100 elderly healthy controls with normal glucose tolerance(normal control group). Results G3316A mutation was detected in 4 cases(4.17%) of T2DM and one case(1.04%) in controls,and there was significant difference between two groups(P0.05).T3394C mutation was detected in 3 cases(3.13%) of T2DM and one case(1.04%) in controls,and there was significant difference between two groups(P0.05).No cross mutation of G3316A and T3394C was found. Conclusion The mutation of G3316A and T3394C in the mitochondrial DNA ND-1 gene could be related to the occurrence of T2DM in the elderly.
