摘  要:目的研究中期因子(midkine,MK)在乳腺癌组织中的表达及其与肿瘤微血管生成的关系,并探讨其临床意义。方法采用免疫组织化学法,对62例乳腺癌组织及32例乳腺良性病变组织(对照)中MK表达情况进行检测;用CD105标记肿瘤新生血管,计数微血管密度(MVD);分析MK、MVD与临床病理特征之间的关系。结果 MK在乳腺癌组织和乳腺良性病变组织中的阳性表达率分别为88.7%(55/62)和12.5%(4/32),差异有统计学意义(P<0.05);乳腺癌组织平均MVD值为21.81±3.53,明显高于乳腺良性病变组织9.52±2.96(P<0.05);乳腺癌组织中MK的表达与MVD呈正相关(r=0.601,P<0.05);MK的表达和MVD计数与乳腺癌患者年龄、肿瘤大小、组织分级等因素无关(P>0.05),但与是否伴有淋巴结转移及临床TNM分期关系密切(P<0.05)。结论 MK在乳腺癌组织中高表达,并与乳腺癌组织微血管生成有关,二者在肿瘤的发生、发展和转移中具有重要作用。Objective To investigate the expression of midkine(MK) and its correlation with microvascular density(MVD) in breast cancer tissues,and to explore their clinical significance in the development of breast carcinoma. Methods Expression of MK was detected in 62 cases of breast cancer tissues and 32 cases of benign breast tissues by immunohistochemistry.MVD was calculated by label-ing new vessels with CD105 antibody.The relationships between MK,MVD and clinicopathological characteristics were analyzed. Results The positive expression rate of MK was 88.7% and 12.5% in breast cancer tissues and benign tissues,respectively(P0.05). The average MVD was 21.81 ± 3.53 in breast cancer tissues,which was significantly higher than that in benign tissues(9.52 ± 2.96,P 0.05).MK and MVD were not associated with age,size of tumor and histological grading(P 0.05),but they were correlated with lymph node metastasis and TNM staging(P 0.05).The expression of MK in breast cancer tissues was positively related to MVD(r =0.601,P 0.05).Conclusion MK is highly expressed in breast cancer tissues,and it is correlated with microvascular angiogenesis.MK and MVD play an important role in the growth,development and metastasis of breast carcinoma.
