纺锤体检测点蛋白BUB1B在胃癌组织中的表达及意义

摘　　要：目的检测胃癌组织中纺锤体检测点蛋白BUB1B的表达,探究BUB1B在胃癌发生中的重要意义。方法采用荧光实时定量PCR检测30例胃癌组织及17例癌旁正常组织中BUB1B mRNA水平表达;免疫组织化学检测BUB1B在胃癌组织及癌旁组织中BUB1B蛋白的表达情况。结果在mRNA水平,BUB1B在胃癌组织中的表达明显高于癌旁正常组织,近(8.875±1.08)倍;免疫组织化学结果显示,胃癌组织中BUB1B的表达明显高于癌旁正常组织。按照评分标准统计显示,癌旁组织BUB1B表达均值为(92.12±12.99),胃癌组织表达均值为(133.39±8.49),两者之间存在明显差异(P<0.05)。且BUB1B表达与TNM分期相关,但与淋巴结转移,年龄,肿瘤大小及有无家族史无明显相关。结论 BUB1B的过表达在胃癌发生发展中起重要作用,为探索肿瘤发生机制及寻找更为有效的治疗提供新的思路。【Objective】To study the expression of spindle checkpoint protein BUB1B and explore the significant value of BUB1B in the initiation of gastric cancer.【Methods】①The mRNA expression levels of BUB1B were compared in tumor tissues of 30 gastric cancer patients with 17 corresponding normal tissues by real-time quantitative PCR.② The immunohistochemistry was used to evaluate of BUB1B protein expression.【Results】① BUB1B mRNA expression levels were higher in gastric cancer than in corresponding normal tissues,nearly 8.875±1.08.② The result of Immunohistochemistry showed BUB1B protein expression were higher in gastric cancer than in corresponding normal tissues,according to the standard scoring,the mean BUB1B protein expression of corresponding normal tissue was 92.12±12.99 and the mean expression of gastric cancer was 133.39±8.49（P〈 0.05）.And the expression of BUB1B is evidently related with TNM stage,but there was no evident correlation with lymph nodes metastasis,age,tumor size and family history.【Conclusions】The overexpression of BUB1B is related with the progression of gastric cancer and provides an new thread to explore the mechanism of cancer initiation and the effective treatments.
