摘 要:目的探讨鼻咽癌组织中梭形癌细胞是否具有上皮-间充质转化(epithelial-mesenchymal transition,EMT)和肿瘤干细胞(cancer stem cells,CSCs)特性。方法以33例鼻咽慢性炎组织作为正常对照,采用免疫组化SP法检测98例鼻咽癌组织梭形癌细胞中E-cadherin、N-cadherin、vimentin、OCT4、SOX2和ALDH1的表达。结果与鼻咽慢性炎组织相比,鼻咽癌组织中梭形癌细胞上皮标志物蛋白E-cadherin明显下调(P<0.001),而间叶标志物蛋白N-cadherin和vimentin明显上调(P均<0.001)。梭形癌细胞中E-cadherin与N-cadherin(P=0.020)和vimentin(P=0.001)的表达呈明显负相关。与鼻咽慢性炎组织相比,鼻咽癌组织中梭形癌细胞中CSCs标志物OCT4、SOX2和ALDH1蛋白明显升高(P均<0.001),二者差异有统计学意义。此外,梭形癌细胞中E-cadherin与OCT4(P=0.029)及ALDH1(P=0.000)的表达均呈负相关。结论鼻咽癌组织中梭形癌细胞具有EMT及CSCs特性,可作为判断转移性鼻咽癌的形态学指标。Purpose To examine whether spindle cells in nasopharyngeal carcinoma (NPC) are associated with epithelial-mesenchy- real transition (EMT) and cancer stem cells (CSCs). Methods Immunohistochemical staining for E-cadherin, N-cadherin, vimen- tin, OCT4, SOX2 and ALDH1 protein was performed on 98 NPC tissues with neoplastic spindle cells, and 33 chronic nasopharyngitis were used as normal controls. Results Compared to normal nasophaEcngeal epithelium, epithelial biomarker E-cadherin expression was down-regulated ( P 〈 0. 001 ) , and the expression of mesenchymal markers N-cadherin ( P 〈 0. 001 ) and vimentin ( P 〈 0. 001 ) was upregulated in the spindle cells. Furthermore, E-cadherin expression was negatively associated with N-cadherin ( P = 0. 020) and vimentin (P =0. 001) in neoplastic spindle cells. Compared with normal nasopharyngeal epithelium, the CSCs markers OCT4 (P 〈 0. 001 ), SOX2 (P 〈0. 001 ) and ALDH1 (P 〈0. 001 ) were overexpressed in the spindle cells. Additionally, E-cadherin expression correlated negatively with OCT4 (P =0. 029) and ALDHI (P =0. 000) in neoplastic spindle cells, respectively. Conclusions For the first time, we demonstrate that neoplastic spindle cells in human NPC are associated with EMT and CSCs, and nfight be used as valuable morphological markers for metastasic NPC.
