摘 要:目的探讨腺病毒介导NK4基因联合单核细胞趋化蛋白1(MCP-1)基因在胰腺癌中的共表达及对胰腺癌细胞的影响。方法利用重组腺病毒AD-MCP-1、AD-NK4分别单独和联合感染胰腺癌细胞株SW1990,荧光定量RT-PCR和酶联免疫吸附剂测定(ELISA)法检测NK4和MCP-1的表达情况并观察其对胰腺癌细胞和血管内皮细胞生长的影响。结果当感染复数(MOI)为60efu/细胞时,SW1990细胞的感染效率接近100%,相应基因mRNA表达在AD-MCP-1组、AD-NK4组和联合组中感染后第5天达到最高,但是联合组中的MCP-1和NK4的表达水平要低于单一感染(P<0.05)。相对于其他各组,联合感染后胰腺癌细胞的相对生长率没有明显的改变。AD-NK4组和联合组的上清均有抑制血管内皮细胞生长的作用,但两组比较差异无统计学意义。结论腺病毒介导NK4联合MCP-1基因感染胰腺癌SW1990细胞没有抑制作用,但NK4能够抑制血管内皮细胞的生长从而发挥抑癌作用。Objective To investigate the effect of the adenovirus-mediated NK4 and MCP-1 gene co-expression in pancreatic cancer cell in vitro.Methods NK4 gene and MCP-1 gene were singly and conjointly transfected into pancreatic cancer cells SW1990 by adenovirus vector.The expressions of NK4 and MCP-1 were detected using RT-PCR and ELISA.Then the influence on the growth of pancreatic cancer cells and vascular endothelial cells were observed.Results When MOI was 60 efu/cell,the infection efficiency of SW1990 was 100%.The expression of MCP-1 and NK4 mRNA were the highest in the 5th days in all the three groups.The expressions of MCP-1 and NK4 in conjoint group were lower than either single MCP-1 or single NK4 group.There was no significant difference in the growth curve of all the three and control groups.Both AD-NK4 and conjoint groups can inhibit the growth of vascular endothelial cells,but no significant difference was found between the two groups.Conclusions Adenovirus-mediated NK4 and MCP-1 genes can effectively infect SW1990,and did not show inhibiting effect in vitro.However,NK4 may play the role of tumor suppressor through inhibiting tumor angiogenesis.
