Fgβ-148C/T、Fgβ-455G/A基因多态性与脑梗死患者颈动脉狭窄的相关性观察

摘　　要：目的探讨血浆纤维蛋白原(Fg)水平及其基因Fgβ-148C/T、Fgβ-455G/A多态性与脑梗死患者颈动脉狭窄的相关性。方法脑梗死患者135例,根据有无颈动脉颅外段狭窄及程度分为脑梗死无狭窄组70例、伴轻度狭窄组42例及中重度狭窄组23例,对照组65例。应用凝固法测定各组血浆纤维蛋白原(Fg)水平,多聚酶链反应-限制性内切酶片段长度多态性技术(PCR-RFLP)检测各组Fgβ-148 C/T、Fgβ-455 G/A基因多态性。结果①脑梗死组血浆Fg水平([3.86±0.95)g/L]高于对照组([3.04±0.53)g/L(]P<0.05);中重度颈动脉狭窄组Fg水平(4.71±0.98)g/L高于轻度颈动脉狭窄组([3.98±1.13)g/L]和无颈动脉狭窄组([3.50±0.54)g/L(]均P<0.05)。②脑梗死组T、A等位基因频率(分别为30.7%和24.8%)明显高于对照组(分别为20.8%和18.5%)(均P<0.05),中重度颈动脉狭窄组T、A等位基因频率(分别为47.8%和45.7%)高于轻度颈动脉狭窄组(分别为40.5%和35.7%)和无颈动脉狭窄组(分别为19.3%和11.4%)(均P<0.05)。结论 Fg水平和Fgβ-148CT、Fgβ-455GA基因位点突变与脑梗死患者颈动脉狭窄及其程度相关。【Objective】 To study the relationship between plasma fibrinogen（Fg） level and β-fibrinogen（Fgβ） gene 148C/T and-455A/G polymorphism and carotid artery stenosis in cerebral infarction patients.【Methods】The plasma fibrinogen levels were measured by solidification assays in 135 patients with cerebral infarction and 65 controls matching.The gene polymorphism of Fgβ-148 C/T and Fgβ-455G/A were detected by polymerase chain reaction restriction fragment length polymorphism（PCR-RFLP）.【Results】 The plasma fibrinogen level in cerebral infarction group（3.86 ± 0.95 g/L） was significantly higher than that those in the control group（3.04 ± 0.53 g/L）（P〈 0.05）;the plasma fibrinogen level in the cerebral infarction patients with moderate to severe stenosis group（4.71 ± 0.98 g/L） was significantly higher than that those in the slight stenosis group（3.98 ± 1.13 g/L） and the no stenosis group（3.50 ± 0.54 g/L）（all P〈 0.05）.The frequencies of T,A allelein in cerebral infarction group（30.7%,24.8%） were significantly higher than that those in the control group（20.8%,18.5%）（all P〈 0.05）;The frequencies of T,A allelein in the cerebral infarction patients with moderate to severe stenosis group was significantly higher than that in the slight stenosis group and the no stenosis group（all P〈 0.05）.【Conclusion】 Plasma fibrinogen（Fg） level and Mutation of β-fibrinogen（Fgβ） gene-148 CT and-455AG polymorphism are associated with the cerebral infarction patients with carotid artery stenosis.
