摘  要:目的研究相对低分子质量岩藻聚糖硫酸酯对慢性乙酸胃溃疡的治疗效果。方法采用慢性乙酸胃溃疡大鼠模型,用灌胃的方式给予相对低分子质量岩藻聚糖硫酸酯(LMW fucoidan)40、100和200mg.kg-1 3个剂量,研究了相对低分子质量岩藻聚糖硫酸酯对胃溃疡大鼠的溃疡指标、氧化程度、NO产生以及MPO的影响。结果灌胃给予LMW fucoidan对大鼠的乙酸胃黏膜损伤有一定辅助治疗作用,与模型组相比胃壁溃疡面积有明显减少。同时对胃组织中的抗氧化酶SOD和GSH-Px有很好的保护作用,显著抑制脂质过氧化产物MDA的产生。与模型组比较,LMW fucoidan组大鼠的胃组织中NO的含量显著上升,MPO的活性显著下降。结论相对低分子质量岩藻聚糖硫酸酯对胃溃疡大鼠的辅助治疗作用与其体内抗氧化活性有关,并通过调节NO产生和炎症细胞的地浸润辅助慢性乙酸胃溃疡的愈合。Objective To investigate the therapeutic effect of relative low mass weight fucoidan from Lam-inaria japonica on pi-deficient rats with gastric ulcers induced by acetic acid. Methods Relative low mass weight fucoidan fracions (LMW fucoidan) were obtained by radical depolymerization and ultrafil-tration of a crude fucoidan from Larninaria japonica. The effects of LMW fucoidan on the acetic acid-induced gastric ulcer rats were evaluated by measuring ulcer bleeding, ulcer index, SOD, GSH-Px and MPO activities, MDA and NO contents and histological characteristics of each rat. Results Compared with the model group, the administration of 100 and 200 mg kg^-1 LMW fucoidan from Laminaria ja-ponica could reduce-bleeding area and ulcer index. The SOD and GSH-Px activities were effectively protected and MDA content was decreased by administration of LMW fucoidan. MPO activity was de-creased and NO content was promoted in the gastric tissues. Conclusion Results showed LMW fucoidan exhibited good therapeutic effects on acetic acid-induced gastric ulcer through its good antioxidant and anti-inflammation activities.
