侵染藠头的大蒜潜隐病毒分子鉴定及部分序列分析

摘  要:藠头(Allium chinenseG.Don),为百合科葱属鳞球茎多年生植物,是一种高产、高营养和具有食疗作用的蔬菜类作物,也常作为中药材加以利用和研究。国内外学者对发生在葱属植物上的病毒病原鉴定和分类做了深入研究,但针对蓖头病害的发生特点和病原鉴定研究不多,自Chen等u0在我国发病野生蓖头样品中发现2种病毒即蓖头花叶病毒(Scallion mosaic virus,ScaMV)和蘸头x病毒(Scallion virus X,ScaVX)后,国内外没有更多有关蓖头病毒病的研究报道。Four Alliurn chinense (G. Don)samples showing viral symptoms were collected from Wuhan city of Hubei Province in China, and subjected to identifying the viruses in the genus Carlavirus by RT-PCR using the degenerate primers pCar-1 ( + ) and M4. Nine clones of amplified products were sequenced and analyzed. The result showed that the amplified fragment covering the 3'terminal genomic RNA was about 1 808 -1 812 nt in length except for poly ( A), and shared a similar genomic organization with that of members in the genus Carla- virus. Sequence alignment showed that they shared similarities of 77% -93% within themselves and high simi- larities ranging from 75% to 82% with that of Garlic latent virus (GLV) isolates. In the phylogenetic tree ba- sing on coat protein sequences, the nine clones clustered into four major groups along with other GLV isolates. This is the first report of GLV infecting A. chinense in China. The obtained results will contribute to the further investigation and management of the viral diseases of A. chinense plants.
