摘 要:目的探究黄酮类化合物槲皮素对大鼠胶质瘤C6细胞侵袭、迁移、增殖及其细胞周期的影响。方法将C6胶质瘤细胞分成Q0、Q50、Q100和Q1504个组;Q0组加适量二甲亚砜处理,Q50、Q100和Q150组槲皮素的浓度分别为50、100和150μmol/L。分别通过transwell体外侵袭实验,transwell体外迁移实验,Click-iT Edu test技术和流式细胞术检测槲皮素对其侵袭迁移,增殖能力及其细胞周期的影响。结果槲皮素处理36h后,Q50、Q100和Q150组的侵袭细胞比率分别为45.81%、13.63%和4.63%;与对照组(100%)相比,其侵袭细胞比率显著降低(P均<0.05)。槲皮素处理12h后,Q50、Q100和Q150的迁移细胞比率分别为:57.98%、33.60%和11.79%;与对照组(100%)相比,其迁移细胞比率显著降低(P均<0.05)。槲皮素处理24h后,Q0、Q50、Q100和Q150组的增殖细胞百分比分别为45.37%、21.49%、7.30%和1.12%;G0/Gl期C6细胞所占比例分别为60.86%、52.11%、35.64%、39.63%,S期分别为34.39%、40.86%、63.48%、60.36%,G2/M期分别为4.76%、6.87%、0.88%、0.11%;Q50、Q100、Q150组的G0/Gl、S、G2/M期细胞比例与Q0组比较,具有显著的统计学差异(P均<0.05);结论研究发现,槲皮素能明显降低胶质瘤细胞的侵袭迁移能力;并可通过阻滞C6细胞的细胞周期进程来抑制其增殖。Objective To study the effects of quercetin on invasion, migration, proliferation and cell cycle of rat glioma C6 cells. Methods The glioma C6 cells were divided into four groups which were Q0, Q50, Ql00 and Q150 group. The Q50, Ql00 and Ql50 group were respectively treated with 50, 100, 150μmol/L quercetin, while the Qo group were treated with DMSO. The influence of quereetin on invasion, migration, proliferation of glioma C6 cells and the change of its cell cycle were detected by transwell in vitro invasion and migra- tion experiment, Click - iT Edu test and Flow cytometry respectively. Results After the treatment for 36h, the invading cell percentage in group Q50 ,Q100 and Q150 were 45. 81% , 13.63% and 4.63% respectively. Compared with Qo group (100%), they significantly decreased ( all P 〈 0.05 ). After treatment for 12h, the migrating cell percentage in group Q50, Q100 and Ql50 were 57.98% , 33.60% and I 1.79% respectively. Compared with Q0 group ( 100% ) , they significantly decreased ( all P 〈 0.05). After treated with quereetin for 24h, the proliferation ratio in Q0, Q50, Q100 and Q150 group were 45.37% , 21.49% , 7.30% and 1.12%. The percentage of C6 cells in the phase Go/G1 were 60.86% , 52.11% , 35.64% and 9.63% , in the phase S were 34.39% , 40.86% , 63.48% and 60.36% , in the phase G2/M were 4.76% , 6.87% , 0.88% and 0.11%. There were statistically significant differences between group Q50, Q100, Q150 and group Q0 in the phase G0/G1 , S, G2/M ( all P 〈 0.05). Conclusion Quercetin could significantly inhibit the invasion and migration of glioma C6 cells and its proliferation by blocking the cell cycle progression.
