摘  要:目的:观察在低O2高CO2条件下,不同剂量的三七皂苷单体Rg1对肺动脉平滑肌细胞(PASMCs)细胞外信号调节激酶(ERK1/2)表达的影响。方法:分离并纯化SD大鼠PASMCs,随机分6组:常氧对照组(N组)、低O2高CO2组(H组)、DMSO对照组(HD组)、Rg1低、中、高剂量干预组(RgL组、RgM组、RgH组)。分别以Western blot和RT-PCR法检测磷酸化ERK(p-ERK)蛋白和ERK1及ERK2 mRNA的表达水平。结果:与N组相比,HD组p-ERK蛋白和ERK1、ERK2 mRNA表达水平明显高于N组(P<0.01);RgL、RgM、RgH组在不同程度上抑制了p-ERK蛋白和ERK1、ERK2 mRNA的表达(P<0.01),其中三七皂苷单体Rg1以40mg/L的浓度为佳。结论:三七皂苷单体Rg1可下调低O2高CO2条件下大鼠PASMCs ERK1/2的表达,这可能是Rg1减轻低O2高CO2性肺动脉收缩(HHPV)的机制之一。Objective: To investigate the effect of notoginsenoside Rg1 with various concentrations on extracellular signal regulated kinase (ERK1/2) signaling pathway in pulmonary arterial smooth muscle cells (PASMCs) under hypoxia and hypercapnia condition. Methods: PASMCs were primarily cultured, and randomly divided into six groups: SD rat under normoxic condition as control group (N group), hypoxia hypercapnia group (H group), DMSO control group (HD group), Rgl-treatment groups (RgL, RgM and RgH group). Western-blot and RT-PCR were used to determine the expression of p-ERK protein and the expression of ERK1 and ERK2 mRNA, respectively. Results: Western-blot and RT-PCR analysis indicated that the expression of p-ERK protein, ERK1 and ERK2 mRNA were significantly higher in HD group than that in N group (P〈0.01). Whereas, the level of ERK1/2 mRNA was markedly lower in Rgl-treatment groups than that in HD group (P〈0.01), and the optimal concentration of Rg 1 is 40 mg/L. Conclusion: Notoginsenoside Rg 1 can attenuate HHPV, which is related to the downregulation of expression of ERK1/2.
