β-连环蛋白和环氧化酶-2在舌癌中的表达及其相关性

摘　　要：目的研究舌癌组织中β-连环蛋白(β-catenin)、环氧化酶-2(Cox-2)的表达及相关性。方法应用免疫组织化学SP法检测β-cate-nin、Cox-2在60例舌癌和30例正常舌黏膜中的表达情况,分析它们与舌癌临床病理指征的关系及二者之间的相关性。结果β-catenin在正常舌黏膜中均正常表达,Cox-2在正常舌黏膜均不表达(P<0.01);β-catenin在60例舌癌中的异常表达率为72%(45/60),β-catenin异位表达与肿瘤TNM分期以及有无淋巴结转移密切相关(P<0.01)。Cox-2在60例舌癌中阳性表达率为80%(48/60),低分化舌癌中Cox-2的表达率高于高中分化舌癌(P<0.01),伴有淋巴结转移的舌癌中Cox-2表达率高于与无淋巴结转移的表达率(P<0.01)。Cox-2在舌癌中阳性表达与与肿瘤分化程度、TNM分期以及有无淋巴结转移密切相关(P<0.01)。Cox-2、β-catenin在舌癌中的表达呈正相关关系(r=0.764,P<0.01)。结论 Cox-2、β-catenin在舌癌的发生发展、浸润及转移过程中起重要作用,Cox-2、β-catenin之间相互作用影响了舌癌的浸润和转移。Objective To observe the expressions of cyclooxygenase-2 and β-catenin in tongue carcinoma,and find out the tumor markers which might be used to predict tumor pathogenesis and metastasis of tongue carcinoma.Methods The expressions of cyclooxygenase-2 and β-catenin in 60 tissue specimens of primary tongue carcinoma and 30 tissue specimens of oral mucosa were detected by S-P method（immunohistochemistry） in order to investigate the relationship between the factors and clinical pathological features of tougne carcinoma.Results In 30 normal cases,β-catenin was positively expressed while Cox-2 was not expressed.In 60 cases of tongue carcinoma,the abnormal expression rate of β-catenin was 72%（45/60）.The expression of abnormal β-catenin was significantly associated with clinical staging（P0.01） and lymph node metastasis（P0.01）.The positive expression rate of Cox-2 was 80%（48/60）.Expression of Cox-2 in poorly differentiated tongue was higher than that of moderately differentiated tongue（P0.01）.Expression of Cox-2 in tongue cancer with lymph node metastasis was higher than that of without lymph node metastasis（P0.01）.The positive expression of Cox-2 was significantly associated with histological grade,clinical staging and lymph node metastasis（P0.01）.Expressions of β-catenin and Cox-2 in tongue was a positive correlation（r=0.764,P0.01）.Conclusions Expressions of Cox-2 and β-catenin play an important role in the development of tongue cancer,degree of differentiation,invasion and metastasis process.The relationship between Cox-2 and β-catenin affects the invasion and metastasis of tongue squamous cell carcinoma.
