摘  要:目的原代培养猪肺动脉内皮细胞,探讨缺氧环境对肺动脉内皮细胞的影响,为肺部疾病提供体外研究模型。方法将猪原代肺动脉内皮细胞分为两组分别置于常氧和低氧环境中培养1、4和7 d,采用免疫荧光法、Western blot及RT-PCR检测各组细胞内皮细胞和间质细胞特异性标志蛋白及mRNA表达。结果常氧组肺动脉内皮细胞呈现典型的鹅卵石样排列,低氧培养的内皮细胞出现长梭形外观。免疫荧光法观察发现低氧培养的内皮细胞开始表达间质细胞抗原α-SMA,且随着低氧时间的延长,α-SMA表达量逐渐增多(P<0.01)。Western blot结果显示与常氧组相比,低氧组内皮细胞VE-cadhrein蛋白表达逐渐下降,α-SMA蛋白表达逐渐上升,RT-PCR结果与此一致。结论低氧促进内皮细胞向间质细胞转化,可能参与体内多种疾病的发生。Objective To examine the effects of hypoxia on endotheliat-to-mesenchymal transition of porcine pulmonary arterial endothelial cells (PAECs). Methods The porcine PAECs were divided into a normoxia group and a hypoxia group. The cells in two groups were exposed to normoxic or hypoxic condition for 1,4, and 7 days respectively. The immunofluorescence, Western blot and RT-PCR were used to detect the protein and mRNA expressions of VE-cadherin and α-SMA. Results The porcine primary PAECs formed typical monolayer of cobblestone appearance on normoxia condition, and had a spindle-shaped appearance on hypoxia condition. Immunofluorescence results showed that these PAECs expressed mesenchymal cells specific marker of α-SMA. With the hypoxic time prolongation, the ratio of transdifferentiated smooth muscle like cells from PAECs was gradually increased ( P 〈 O. 01 ). Western blot assay demonstrated that the expression level of VE-cadherin protein and mRNA was reduced gradually, but the expression level of α-SMA protein and mRNA was increased. Conclusion Hypoxia can induce endothelial-to-mesenchymal transition, which may be involved in the development of a variety of diseases.
