摘 要:目的研究丁香酚对大鼠局灶性脑缺血/再灌注损伤的神经保护作用机制。方法参照Longa报道的方法[1]制备大鼠局灶性脑缺血/再灌注模型。48只SD♂大鼠随机分为假手术组、模型组(MCAO)、丁香酚组(MCAO+丁香酚)。缺血/再灌注后12 h进行行为学评分,22 h TTC染色测定梗死面积,72 h HE染色检测脑组织病理变化,并检测大鼠脑组织匀浆中谷胱甘肽(GSH)、丙二醛(MDA)、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)的变化,应用蛋白质印迹检测大鼠脑组织中NF-κB的表达。结果丁香酚能明显改善大鼠的神经行为,缩小梗死面积,使脑组织病理改变减轻,GSH、SOD、CAT活性明显升高,MDA含量明显降低,下调NF-κB的表达。结论丁香酚可能通过抑制氧化损伤和炎症来减轻大鼠脑缺血/再灌注性损伤。Aim To investigate the neuroprotective effects of eugenol on cerebral ischemia / reperfusion injury in rats. Methods According to the method reported by Longa, rat focal cerebral ischemia/reperfusion model was prepared. 48 male SD rats were randomly divided into sham group, model group and eugenol group. Behavioral score was determin after 12 h; TFC staining was used to determinate infarction area after 22 h; HE staining was adopted to detect pathological change in injury brain tissue after 72 h. At the same time, the levels of GSH, SOD, MDA and CAT in brain tisshe were treated, the NF-κB protein expressions were detected by Western blot. Results Euge-nol could significantly improve the rats' neural behavior, reduce cerebral infarction area, and relieve the injury of brain tissue. It increased the activity of GSH, SOD and CAT and decreased the contents of MDA. It also reduced the expression of NF-κB. Conclusion Eugeno] reduces cerebral ischemic injury by suppression of oxidative injury and inflammation.
