摘 要:目的探讨MACC1和C—MET蛋白在肝细胞癌组织中的表达及意义。方法采用免疫组化和Western blot检测51例肝细胞癌组织、相应癌旁组织和13例正常肝脏组织中MACC1和C—MET蛋白的表达情况,同时对患者进行随访,并分析患者生存率与临床病理资料间的关系。结果MACC1和C—MET蛋白在肝细胞癌组织中总的阳性表达率分别为80.4%和76.5%,相对表达量分别为0.645±0.047和0.504±0.023,均明显高于相应癌旁组织及正常肝脏组织(分别F=173.308,252.817,均P=0.000),且两者表达呈正相关(r=0.870,P=0.001);MACC1和C—MET蛋白表达阳性患者的3年生存率明显低于阴性患者(分别Х^2=3.934,4.439,均P〈0.05),两者的阳性率及表达量均与肝细胞癌的TNM分期、门脉癌栓、病理分型及包膜侵犯显著相关(P〈0.05)。结论MACC1和C—MET的异常表达与肝细胞癌的恶性进展相关,MACC1有作为进展期肝癌预后的独立分子指标和肝癌治疗新靶点的潜在价值。Objective To explore the expression and clinical relevance of metastasis-associated colon cancer-1 (MACC1) and C-MET proteins in hepatoeellular carcinoma (HCC) tissue. Methods The expressions of MACC1 and C-MET were detected in 51 specimens of HCC and paraneoplastic liver tissue, normal liver tissue in 13 healthy cases using immunohistochemistry and Western blotting. The correlations of the expressions of MACC1 and C-MET proteins were evaluated, survival rates were observed ,the relationship between the expression of MACC1, C-MET proteins and the clinieopathologic features of HCC were analyzed. Results The positive rate of MACC1 and C-MET proteins was 80. 4% and 76. 5% in HCC tissue, the relative expressions were 0. 645 ±0. 047 and 0. 504 ±0. 023 respectively, which was significantly different from those in paraneoplastic liver tissue and normal liver tissue (respectively F = 173. 308,252. 817, all P = 0. 000). The survival analysis showed that the three-year survival rate in patients with positive MACC1 and C-MET expressions was significantly lower than that in patients with negative expressions ( respectively Х^2 = 3. 934,4. 439, all P 〈 0. 05 ), the positive rate and relative expressions of MACC1 and C-MET were significantly correlated with TNM stage, portal vein cancer thrombus and pathology typing ( P 〈 0. 05 ). Conclusions The expression of MACC1 and C-MET is associated with the malignant progression of HCC. MACC1 may serve as a independent prognostic factor for advanced HCC and a possible therapy target for the treatment of HCC.
