摘　　要：目的探讨RNA结合蛋白La在宫颈癌组织中的表达及在宫颈癌发生、发展过程中的作用。方法采用免疫组化染色技术分别测定La蛋白在宫颈癌和正常宫颈组织中的表达情况;采用RNA干扰技术沉默La基因在宫颈癌细胞株HeLa的表达,并通过G418筛选,构建稳定表达的HeLa的La干扰克隆。然后使用HeLa-shLa和HeLa-shNC分别进行细胞增殖、体外肿瘤球形成实验,细胞周期检测和Western blot等。结果宫颈癌组织中La蛋白的表达61%明显高于正常宫颈组织9%,差异有统计学意义(P<0.05);且干扰La后,HeLa细胞的增殖能力及肿瘤球形成率(shNC组:17.1%±1.92%,shLa组:6.3%±0.45%)均出现降低,与载体对照组相比,差异具有统计学意义(P<0.05)。此外La蛋白干扰后,HeLa细胞发生明显的G0/G1期阻滞,且细胞内CyclinD1的表达降低。结论 RNA结合蛋白La对宫颈癌的形成具有促进作用,可能在宫颈癌的发生、发展中起重要作用。Objetive To investigate the expression of La protein in cervical cancer tissues and explore its role in the occurrence and progression of cervical cancer. Methods The expression of La protein in cervical cancer and normal cervical tissues was detected by immunohistochemical staining. RNA interference technology was used to silence La protein expression in HeLa cells and the changes in cell proliferation, tumor sphere formation and cell cycles were investigated. Results The expression of La protein was significantly higher in cervical cancer tissues than in normal cervical tissues （61% vs 9%, P0.05）. Silencing La protein expression in HeLa cells caused significantly reduced the cell proliferation and lowered the tumor sphere formation rate from the control level of （17.1±1.92）% to （6.3±0.45）% （P0.05）, resulting also in G0/G1 cell cycle arrest and reduced cyclin D1 protein expression. Conclusion The RNA binding protein La can promote the development of cervical cancer and may play a critical role in the carcinogenesis and progression of cervical cancer.
