摘 要:目的探讨金属硫蛋白(MT)对大鼠缺血再灌注(I/R)心肌细胞凋亡及核因子κB(NF-κB)表达水平的影响。方法 32只SD大鼠随机分成实验组、对照组,每组16只。分别予硫酸锌(ZnSO4)和蒸馏水预处理。预处理后构建Langendorff离体心脏I/R动物模型,动态观测缺血前及再灌注期左心室发展压(LVDP)、左心室压力最大上升和下降速率(±dp/dtmax)、冠状动脉流出量(CF);测定冠状动脉流出液肌酸磷酸激酶MB同工酶(CK-MB)的漏出率;Western印迹法检测金属硫蛋白(MT)表达;TUNEL法检测心肌细胞凋亡;免疫组化法检测NF-κB的活化水平。结果与对照组比较,实验组再灌注期LVDP、±dp/dtmax及CF得到改善(P<0.05);CK-MB的漏出率减少;金属硫蛋白的表达增加;细胞凋亡指数及NF-κB表达减少(均P<0.01)。结论 MT下调NF-κB表达可能与其抑制I/R心肌细胞凋亡有关。Objective To explore the effects of induced metallothionein(MT) on myocardial apoptotic and nuclear factor-kappa B(NF-κB) expression in rat hearts during ischemia/reperfusion.Methods 32 SD rats were divided randomly into the experiment and control groups.Rats had been pretreated with 3.6% ZnSO4 or distilled water before their hearts were separated for Langendorff perfusion and for ischemia/reperfusion.The left ventricular functions(LVDP,±dp/dtmax),coronary artery flows(CF) were observed dynamically before ischemia and during 60-minute reperfusion following 30-minute ischemia.The levels of CK-MB in the coronary artery flow were measured.Apoptosis was measured by TUNEL staining method.MT was assessed by using western blot.NF-κB activation was examined by using immunohistochemistry.Results Compared with control group,the LVDP,±dp/dtmax and CF were greatly improved(P0.05),the level of CK-MB,the apoptotic index and the activation of NF-κB were significantly decreased(P0.01) in experiment group.Conclusions MT can induce down-regulation of NF-κB,which function might inhibit myocardial apoptosis after ischemia/reperfusion in rat.
