摘　　要：目的探讨多发性骨髓瘤(MM)中IL-8基因多态性及硼替佐米(万珂)的干预效果。方法在多种细胞株中找到3个细胞株分别含有IL-8启动子区-251位的三个基因多态性,加入万珂后检测分析各细胞株生存率,并测定各细胞株IL-8水平以及IL-8 mRNA表达水平。结果基因型为A/A的纯合子细胞株IL-8表达量最高,对万珂最敏感,T/T细胞株IL-8表达量最低对万珂最不敏感。结论 MM中IL-8基因存在多态性,且与万珂干预治疗有关。Objective To explore the gene polymorphisms of interleukin-8 in multiple myeloma（MM） and the therapeutic effect of Velcade.Methods Three cell lines were identified from many MM cell lines and contained three polymorphisms for the IL-8 promoter region-251 respectively.Cell survival rate was measured after treatment of Velcade.IL-8 cytokine levels as well as quantification of IL-8 mRNA were tested.Results The efficacy relationship of MM cell lines to Velcade was homozygous A/A heterozygous A/T homozygous T/T,so as the concentrations of IL-8 in the tissue culture fluid and quantitative levels of IL-8 mRNA transcripts.Conclusions Response to therapy with Velcade in MM cell lines is related with gene polymorphisms of IL-8 promoter region.
