摘  要:目的观察在常压7%氧浓度环境下于不同持续时间大鼠小脑浦肯野细胞组织学及其超微结构的急性损伤病理改变。方法 Wistar大鼠120只,随机分为缺氧组和对照组,雌雄各半,每组60只,其中缺氧组大鼠置入常压低氧舱内,氧浓度为7%,于持续缺氧1、3、5、7、10、14 d与相应对照组分别处死,小脑取材、固定、制片,观察其光镜和电镜下的结构变化,并从每只大鼠小脑组织所制的切片中随机选片,于光镜下自桥脑侧计数500个浦肯野细胞,计算变性细胞的发生率。结果从持续缺氧1~7 d过程中,浦肯野细胞的变性程度呈进行性加重,缺氧性改变细胞的数量呈进行性增加;于持续缺氧10~14 d时,缺氧性改变的程度呈进行性减轻趋势,变性细胞的数量亦呈减少趋势。光镜观察、电镜观察及变性细胞发生率三者结果一致。结论在不同持续常压缺氧过程中大鼠小脑浦肯野细胞的变性程度及其数量呈早期进行性加重和增多—后期渐适应恢复的规律性变化。Objective To observe the pathological change of histological and ultrastructural of Purkinjecells in the rat cerebellum after different duration in the normobaric environment of 7% oxygen concentra- tion. Methods Sixty female and 60 male rats were randomly divided into 6 hypoxia groups, namely, 1, 3, 5, 7, 10 d, and 14 d groups, and control group. The rats of hypoxia groups were put into the normo- baric hypoxic cabin where the air oxygen concentration inside was 7%. Cerebellum samples were obtained after 1, 3, 5, 7, lO, and 14 day hypoxia. Histological and ultrasections were prepared and observed un- der light and electron microscopes. Some of microtome sections was picked out from the all microtome sec- tions of every rat randomly. The rate of degenerating cells in 500 Pukinje cells was counted. Results The degree of cell degeneration was aggravated gradually and the number of degenerating cells was gradu- ally increased during the day 1 to day 7, and then the above - mentioned cellular changes were gradually reduced during the day 10 to day 14. The results observed by histology using HE staining, count of degen- erating cells and the ultrastructural observation were consistent. Conclusion There is a certain regularity of the changes of degree and quantity of degenerating Purkinje cells in the cerebellum of rat after different duration of normal pressure hypoxia, i. e. aggravating rapidly within early phase--adapting and recove- ring slowly within late phase
