异丙酚对大鼠脑缺血再灌注后p63各亚型表达的影响

摘　　要：目的探讨异丙酚对大鼠脑缺血再灌注后p63亚型(TAp63、ΔNp63)表达的影响。方法将120只健康SD大鼠随机分为假手术(S)组、脑缺血/再灌注(I/R)组和异丙酚(PP)组,每组40只。分别于术后6 h,1、3、5 d处死三组大鼠,用免疫组化法检测TAp63、ΔNp63的表达,原位末端转移酶标记技术(TUNEL)检测神经元凋亡情况。结果与S组比较,I/R组TAp63表达率于术后6 h,1、3 d逐渐增加,3 d与1、5 d比较,差异有统计学意义(P<0.05);ΔNp63表达率在术后逐渐增加,各时间点比较差异均有统计学意义(P<0.05);PP组TAp63表达较I/R组同时间点明显减少(P<0.05),而ΔNp63表达较I/R同时间点增多(P<0.05)。I/R和PP组1、3、5 d细胞凋亡指数较S组增加(P<0.01);PP组较I/R组同时间点降低(P<0.05),3 d与1、5 d比较,差异有统计学意义(P<0.05)。结论 TAp63表达在I/R后呈先增加后减少趋势,与神经元凋亡趋势基本一致,提示TAp63可能参与了神经元的凋亡;PP具有一定的脑保护作用,其机制可能与下调TAp63、上调ΔNp63表达进而抑制神经元凋亡有关。Objective To discuss the influence of propofol on the expression of p63 isoforms（TAp63,ΔNp63） after cerebral ischemia-reperfusion in rats.Methods 120 healthy SD rats were randomly divided into the sham-operation（S） group,cere bral ischemia/reperfusion（I/R）group and propofol（PP）group,40 cases in each group.The rats of three groups were executed at 6 h,1,3,5 d after operation.The immunohistochemical method was used to determine the expression of TAp63 and ΔNp63;the terminal-deoxynucleoitidyl transferase mediated nick end labeling（TUNEL） method was used to detect the expression of the apoptosis of neurons.Results Compared with the S group,the expression rates of TAp63 at 6 h and on 1,3 d in the I/R group were in creased gradually,and which on 3 d showed statistical differences compared with that on 1,5 d（P0.05）;the ΔNp63 expression rate was gradually increased after operation,and the difference at different timepoints had statistical significance（P0.05）;the TAp63 expression in the PP group was decreased more obviously than that in the I/R group at the same timepoint（P0.05）,but the ΔNp63 expression was increased more obvious than that in the I/R group（P0.05）.The apoptosis indexes on 1,3,5 d in the I/R and PP groups were significantly higher than that in the S group（P0.01）;which in the PP group were lower than that in the I/R group at the same timepoint（P0.05）;the difference between the comparisons on 3 d and 1,5 d had statistical significance（P0.05）.Conclusion The expression of TAp63 isoform after cerebral ischemia/reperfusion shows the trend of firstly increasing and then decreasing,and is consistent with the neuronal apoptosis trend,which suggests that TAp63 may be involved in neuronal apoptosis.Propofol has certain protective effect on the brain,and its mechanism may be related with the downregulation of TAp63 expression,upregulation of ΔNp63 expression and the inhibition of neuronal apoptosis.
