摘　　要：目的:观察NGFIB-β(nerve growth factor-induced gene B-β,Nurr1)在大鼠视网膜神经细胞发育过程中蛋白表达的动态变化及与PCNA的相关关系,探讨NGFIB-β在视网膜发育过程中的表达意义。方法:石蜡包埋组织切片,免疫组织化学双重染色及Western blot。结果:NGFIB-β在视网膜发育过程中的表达呈现了显著的动态变化,NGFIB-β从E18 d开始在大鼠视网膜组织出现少量表达,阳性表达主要在内核层内缘,随着发育的进行阳性细胞数目明显增多,P3～7 d达到高峰,之后随着细胞的成熟阳性表达又逐渐减少,成熟的视网膜组织内仅见少量阳性细胞,与PCNA的对比观察发现,两者表达在不同部位和不同细胞中,NGFIB-β主要表达在已分化的幼稚神经细胞中,在增殖细胞中不表达;PCNA蛋白从E14 d到P7 d为强阳性表达,P9 d时阳性细胞的数目明显减少,成熟的组织中未见阳性表达。结论:NGFIB-β在大鼠视网膜神经细胞从幼稚到成熟的分化过程中可能有重要调控作用。Objective： To observe nerve growth factor-induced gene B-β （NGFIB-β, Nurrl ） protein expression patterns during rat retina development and the correlation with proliferating cell nuclear antigen （PCNA）, and to discuss the sig- nificance of NGFIB-β expression in retinal development of rat. Method： We employed paraffin embedding tissue section, immunohistochemical double staining and Western blot methods. Results： The results revealed dramatic and dynamic changes of NGFIB-β protein expression during retinal development. NGFIB-^- positive cells appeared in small quantities at embryonic day 18, and their number then increased markedly during development. The peak occurred at postnatal days 3-7. As maturation continued, the number of positive cells gradually decreased. Comparative observation of NGFIB-βand PCNA showed that NGFIB-β was confined to differentiate and migrate immature cells, and that it was not present in prolif- erating cells. PCNA showed a prominent positive expression from El4 to P7, and then the positive cells appeared a signif- icantly reduction at P9. Conclusion： These results suggest that NGFIB-β may play a regulatory role during differentiation and maturation of the rat retina.
